Embodiment

[0097] Propionibacterium acnes phage-derived recombinant endolysin and thymol; and the combination of thymol + terpineol Combined synergy.

[0098] Propionibacterium acnes 6919 strain (ATCC, USA) was grown on RCM agar under anaerobic conditions by using glycerol stock. A loop of the culture from the glycerol stock was streaked onto freshly prepared RCM agar plates and incubated anaerobically at 37°C for 4-5 days. After incubation, the culture was removed from the plate and resuspended in saline, then centrifuged at 8000 rpm for 6 min at 4°C. The obtained pellet was resuspended in 10 mM HEPES buffer (pH 7.0). Prepare an OD of 0.5 in HEPES buffer 600 Cultures. A 1:10 dilution of the 0.5OD conditioned culture was used for a contact kill assay, which was equivalent to 10 6 cells / ml.

[0099] The assay was performed in a total volume of 200 μl. 14 μl of purified endolysin (final concentration 100 μg / ml) was used for the assay. Essential oils were freshly prepared at 10% w...

Abstract

The present invention relates to a method and a composition to prevent or treat acne by selective inhibition of P. acnes bacteria. The method comprises treating skin with P. acnes phage-derived endolysins and nucleic acid molecules encoding the same in combination with selective essential oils.

Description

technical field [0001] The present invention relates to a method and composition for preventing or treating acne by selectively inhibiting Propionibacterium acnes (P. acnes) bacteria. The method comprises treating the skin with an endolysin in combination with an essential oil compound. Background technique [0002] Acne, also known as Acne vulgaris, is a common skin condition that affects nearly all teens and adults at some point in their lives. Its etiology is complex and involves abnormal keratinization, hypersebum production, androgen function, bacterial growth, and immune hypersensitivity. Although one or more of the above processes are associated with acne, a trigger and the exact sequence of events leading to the formation of acne lesions is not fully understood. Other factors associated with acne are the presence of free radicals and subsequent oxidative stress leading to cellular damage. Acne has been observed to commonly occur in areas rich in sebaceous glands s...
