Embodiment 1

[0041] Molecular characteristics of the astragalus lectin protein prepared in Example 1

[0042] After gel filtration analysis and SDS-PAGE electrophoresis analysis, its molecular weight is 61.1kDa, and the molecular weight of a single subunit is 29.6kDa. It is a homodimer composed of two subunits in the natural state, as shown in the attached figure 1 shown. After SDS-PAGE electrophoresis, it was identified by periodic acid-Schiff (PAS) reagent method, and there were obvious pink bands at the corresponding positions, indicating that the lectin protein was a glycoprotein.

[0043] Amino acid composition analysis showed that the protein was rich in glycine (Gly, 16.0%), serine (Ser, 11.0%), aspartic acid (Asp, 9.9%) and leucine (Leu, 9.6%), and cysteine acid, histidine and methionine were not detected. The composition of N-terminal amino acid residues was determined on Applied Biosystems Procise-PROCISE by Edman degradation method. The N-terminal amino acid sequence is: ESGI...

Abstract

The present invention provides an agglutinant protein obtained by separating plant astragalus root and purifying its extract and its preparation method. In the concrete, said invention uses the astragalus root as raw material, and utilizes ammonium sulfate with different saturations to make fractional precipitation and multistep ion exchange column chromatography so as to obtain astragalus agglutinant protein which can be used for blood agglutination and has the obvious antifungal action.

Description

technical field [0001] The invention belongs to the field of plant protein, and more specifically relates to isolating a lectin protein from Astragalus membranaceus and a preparation method thereof. Background technique [0002] Chinese herbal medicine is mainly used according to the active ingredients contained in it. In order to make full use of resources and explore new active ingredients, protein ingredients have gradually attracted attention. In recent years, many proteins with biological activity have been found in various Chinese herbal medicines. Separation and purification of active protein components in Chinese herbal medicines, in order to find some new proteins or peptides with clinical application value, has become an important direction for many researchers at home and abroad. [0003] Plant lectin (lectin) is a kind of binding specific carbohydrate protein, also known as hemagglutinin (haemagglutinin) or agglutinin (agglutinin), is a non-immune origin, non-en...
