Abstract

An integrated lab-on-a-chip diagnostic system for carrying out a sample preparation process on a fluid sample containing cells and/or particles, the system comprising: (a) an inlet for a fluid sample; (b) a lysis unit for lysis of cells and/or particles contained in the fluid sample; (c) a nucleic acid extraction unit for extraction of nucleic acids from the cells and/or particles contained in the fluid sample; (d) a reservoir containing a lysis fluid; (e) a reservoir containing an eluent for removing nucleic acids collected in the nucleic acid extraction unit; wherein the sample inlet is in fluid communication with the lysis unit, an optional valve being present to control the flow of fluid therebetween; wherein the lysis unit is in fluid communication with the nucleic acid extraction unit, an optional valve being present to control the flow of fluid therebetween; wherein the reservoir containing the lysis fluid is in fluid communication with the lysis unit, an optional valve being present to control the flow of fluid therebetween; and wherein the reservoir containing the eluent is in fluid communication with the nucleic acid extraction unit, an optional valve being present to control the flow of fluid therebetween.

Description

technical field [0001] The present invention relates to the extraction of nucleic acid (NA), in particular, the present invention relates to an integrated lab-on-a-chip diagnostic system for combined extraction and concentration of NA. The system can be used to perform NA sequence amplification and detection processes on fluid samples containing cells. Background technique [0002] It is of considerable interest to develop simplified analytical systems for the detection of biomolecules which allow unskilled users to carry out complex analytical procedures without undue error. Furthermore, it is of great interest to develop an analytical system that contains a minimum of liquid reagent handling and automates the analytical procedure with minimal user intervention, and is also preferably miniaturized such that Provides a convenient system for point-of-care testing. This is especially relevant in the field of healthcare, especially diagnostics, where there is a growing need f...
