Embodiment Construction

[0030] The compositions of the present invention combine an RNAIII-inhibiting peptide with a polycationic antimicrobial peptide capable of binding and neutralizing lipidic and polyanionic components of the bacterial envelope, such as LTA or LPS. The antimicrobial peptide may be cathelicidin. The RNAIII-inhibitory peptides of the invention are generally peptides capable of inhibiting RNAIII activity, reducing TRAP phosphorylation, inhibiting cytokine or NO production in in vitro models, or exhibiting related activities. Recognizing the importance of cell wall components and exotoxin production in the pathology of bacterial sepsis, the compositions of the present invention are advantageously used in methods of treating bacterial sepsis or similar conditions in which the bacterial pathology is related to the bacterial outer cell envelope The lipids are associated with polyanionic components such as LTA or LPS.

Abstract

The present composition combines an RNAIII-inhibiting peptide (RIP) with an antimicrobial peptide, such as a cathelicidin, that is capable of binding and neutralizing lipidic and polyanionic components of bacterial cell envelope. In another embodiment, the RIP is combined with an antibiotic, with or without an antimicrobial peptide. The present composition is advantageously used in a method of treatment of bacterial sepsis.

Description

[0001] Cross References to Related Applications [0002] This application claims priority to US Provisional Patent Application Serial No. 60 / 668,132, filed April 4, 2005, which is hereby incorporated by reference in its entirety. technical field [0003] The present application mainly relates to pharmacological compositions and methods for treating bacterial infections or reducing the risk thereof, in particular to compositions comprising RNAIII-inhibiting peptides and antimicrobial peptides and / or antibiotics such as aminoglycosides, beta-lactams , cephalosporin (caphalosoprin) or vancomycin (vancomycin). Background technique [0004] sepsis [0005] Despite improvements in antimicrobials and better supportive care, sepsis remains the leading cause of death. Sepsis is associated with systemic inflammation, circulatory failure and multiple organ dysfunction syndrome (MODS). Both Gram-positive organisms, such as Staphylococcus aureus, and Gram-negative bacteria can caus...
