Embodiment 1

[0029] Take 0.5 kg of hematoxylin and use 80% ethanol which is 6 times its weight to reflux and extract once, each time for 1 hour. The extract was decompressed, concentrated at a temperature of 45°C, recovered ethanol, dried, and weighed. Add methanol to dissolve, stir into the polyamide column, and then use 4 times column volume of water and 4 times column volume of 90% ethanol to elute respectively, and collect the latter part of the eluate. The collected eluate was concentrated under reduced pressure at a temperature of 30° C., and dried under reduced pressure to obtain brazilianin. Measured by high performance liquid chromatography, the brazilian hematoxylin content is about 80%.

Embodiment 2

[0031] Get 0.5 kg of hematoxylin and use 15 times of the weight of the material to extract 2 times with 80% ethanol under reflux, each time for 3 hours. Concentrate the extract to recover ethanol, mix it with silica gel and pack it into a column, elute with 6 times column volume of ethanol-chloroform (20:80), and collect the eluate. The collected eluate was concentrated under reduced pressure at 60° C., and the precipitate was dried under normal pressure to obtain brazilian hematoxylin. The brazilian hematoxylin is determined by high performance liquid chromatography, and the brazilian hematoxylin content is about 90%.

Embodiment 3

[0033] Get 0.5 kg of hematoxylin and extract with 95% ethanol which is 10 times of its weight under reflux, at a reflux temperature of 80° C., and extract 3 times, each time for 1 hour. Concentrate the extract under reduced pressure at 50°C, recover ethanol, add water, extract with petroleum ether, and concentrate the extract under normal pressure to obtain brazilian hematoxylin. The brazilian hematoxylin is determined by high performance liquid chromatography, and the brazilian hematoxylin content is about 92%.

[0034] In the application of medicaments, brazilian hematoxylin is prepared into medicaments by conventional preparation process either alone or in a compound. The medicament can be various dosage forms that can be used clinically.

[0035] Determination of Brazilian hematoxylin, using high performance liquid chromatography to determine its content.

Abstract

The invention discloses novel use for brasilein, which relates to the novel use of the brasilein with brazilwood as raw material in the medical aspect, in particular to an application of the brasilein in the preparation of medicines used to treat cerebral ischemia or cerebral hemorrhage, brain tissue peroxide increase and brain function damage diseases caused by the brain tissue peroxide increase. As shown in experimental research, the brasilein has the effects on resisting oxidation, protecting brain neurons, enhancing cerebral blood flow, and suppressing the activity of erythrocyte membrane Na-K-ATP enzyme. The brasilein has an apparent effect on brain issue damage caused by cerebral ischemia reperfusion. When the diseases are treated, the brasilein, taken as an active ingredient, is independently used or matched with other medicines to prepare various medicines with different preparation formulations, and the medicines can be clinically used.

Description

[0001] This application is a divisional application with the patent application number of 200410004758.9 and the title of "New Application of Brazilian Hematoxylin". technical field [0002] The invention relates to a new application of brazilian hematoxylin extracted from hematoxylin as a raw material in medicine, and belongs to the technical field of traditional Chinese medicines. Background technique [0003] Sumac is the dried heartwood of the leguminous plant Caesalpinia sappan L. Distributed in my country's Guangxi, Guangdong, Taiwan, Guizhou, Yunnan, Sichuan and other places. Available all year round. Remove the skin and sapwood, take the heartwood and dry it in the sun. Contains brasilin, brasilein and other ingredients. Brazilian hematoxylin (C 16 h 12 o 5 ) has the following structural formula: [0004] [0005] Physical and chemical characteristics: [0006] Red-brown crystals (MeOH), mp 260-265°C (dec.), [a] D 20 -1008 (MeOH; c 0.6).
