Problems solved by technology

The discovery and utilization of the semi-dwarf gene in the 1950s and 1960s increased the rice yield by 20% to 30%, realizing the first leap in rice yield; Great breakthroughs have been made in the utilization of advantages, but for a period of time, the problem of single dwarf stem genes in rice breeding in my country has become more and more serious, and the innovation of dwarf stem resources in rice breeding has been relatively slow, which has become an important factor affecting the further increase in the yield of rice breeding varieties one
At present, 37 CSL genes have been found in the rice genome by using bioinformatics methods (Hazenet al. 2002), but the definite functions of these genes lack specific experimental evidence, and only stay on the prediction based on the gene sequence

Embodiment 1

[0023] 1. Materials and methods

[0024] 1. Materials

[0025] Mutants were derived from the A c / Ds insertion mutant library constructed in our laboratory (Luan et al., 2008) 15 , for the background of Oryza sativa L.ssp.Japonica cv.Nipponbare. The mutants were crossed with Oryza sativa L. ssp. Indica cv. Nipponbare to produce the F1 generation, and the F1 generation was self-crossed to generate the F2 generation segregation population.

[0026] All materials were planted in the experimental field of China Rice Research Institute.

[0027] 2. Method

[0028] 2.1 Micro-extraction of rice genomic DNA

[0029] Extracted by simple CTAB method. Specific steps are as follows:

[0030] 1) Take about 300mg of fresh young rice leaves, freeze them with liquid nitrogen, grind them into powder, and quickly transfer them to a 1.5mL Enpdoff tube;

[0031] 2) Add 650 μL of CTAB extraction buffer (100mM Tris-HCl pH 8.0, 20mM EDTA, 1.4M NaCl, 2.0% CTAB, 1% PVP) preheated at 65°C, bathe ...

Embodiment 2

[0138] 3. Prediction of the application prospect of the cloned target gene

[0139] Polysaccharides play an important role in the development of plant cells, mainly including cellulose, hemicellulose and pectin. On the one hand, these polysaccharides are part of cell morphogenesis; on the other hand, they play important roles in the environment and physiology of cell survival. The synthesis and metabolic pathways of these polysaccharides are very complex. It is currently known that glycosyl transferases (GTs) play an important role in the synthesis of polysaccharides. The gene encoding this enzyme is a superfamily of which CSLDs are among them. A subfamily of CSLDs, in existing reports, the function of CSLDs subfamily is not very clear, in Arabidopsis, it is related to the synthesis of xylan and homopolygalacturonic acid, which affects the development of stems. OsCSLD1 was reported to be involved in the development of root hairs in rice. In this study, a dwarf leaf-rolling m...

Abstract

The invention relates to rice dwarf straw leaf roll mutant (cd1) gene having the nucleotide sequence SEQ ID No.1, the nucleotide sequence of the corresponding wild type OsCD1 gene is SEQ ID No.2. In the invention, a dwarf straw leaf roll mutant is discovered in an established insert mutant database, the gene (cd1) thereof discloses that the mutant character is controlled by CSLD subfamily genes. In the research, the character shown by the mutant seriously influences the growth and development of rice, the shape construction of the plant is suppressed, the chlorophyll content is lowered, the ripening rate decreases greatly, the seeds are not full, and evolution of agronomic trait is limited; therefore, the inventive rice dwarf straw leaf roll mutant (cd1) gene provides reliable theoretical basis for drought combat, disease resistance and lodging resistance, particularly the invention can change plant shape by using gene engineering method, regulate and control the shape of plant, and has important utilization value in gardening ornamental plants.

Description

[0001] This patent is funded by the National Natural Science Foundation of China (No. 30871328) and the National Transgenic Major Project (2008ZX08009-003). technical field [0002] The invention belongs to the technical field of improving the overall growth and development of rice, and relates to a curl leaf and dwarf 1, cd1 mutant (curl leaf and dwarf 1, cd1) gene that affects the overall growth and development of rice and its application. Background technique [0003] Rice is the main food crop in China and plays a pivotal role in my country's agricultural production and food security. With the increasing demand for food and the continuous reduction of arable land, increasing rice yield has been paid more and more attention, and high and ultra-high yield has always been the theme of rice research. The discovery and utilization of the semi-dwarf gene in the 1950s and 1960s increased rice yield by 20% to 30%, realizing the first leap in rice yield; in the 1970s and 1980s, t...
