Embodiment Construction

[0014] The method experimental device of the present invention is as figure 1 shown. The THz-TDS system uses a titanium-doped sapphire (Ti: sapphire) self-mode-locked femtosecond laser (LM Labs, USA), with a working center wavelength of 800nm, a pulse width of 25fs, a repetition rate of 88MHz, and a pulse interval of 11.4ns. The high-power green laser Verdi-V6 from Coherent Corporation of the United States was used as the pump source of the titanium-doped sapphire femtosecond laser. A broadband photoconductive switch is used as the transmitter and receiver of terahertz waves. There are 4 elliptical mirrors to form an 8-F confocal geometric optical system. The beam waist spot diameter is 3.5mm. The spectral bandwidth is 4.5THz (3mm~67μm), and the signal-to-noise ratio is greater than 30000:1. The optical delay line of the precise micro-displacement platform driven by a stepping motor is used to control the detection light pulses (femtosecond pulses) arriving at different ti...

Abstract

The invention relates to a method for quickly detecting amyloid protein fiber by thzTHz time-domain light spectrum, which is used for detecting misfolded or denatured protein. The method comprises the following steps: 1. measuring the thzTHz time-domain light spectrum of a series of normal protein corresponding to protein to be detected, and building establishing a light spectrum database; 2). measuring the thzTHz time-domain light spectrum of the protein sample to be detected; and 3). according to the light spectrum database, comparing the light spectrum data of normal protein, and analyzing whether the protein sample to be detected forms amyloid protein fiber. The method has simple process, needs small amount of samples, has short detection time and can perform lossless high-sensitivity detection to amyloid protein fiber.

Description

【Technical field】 [0001] The invention belongs to the technical field of biomedical molecular detection, and the technology involved is a method for rapid and nondestructive detection of amyloid protein by using terahertz time-domain spectroscopy technology. 【Background technique】 [0002] Amyloid is an abnormal fiber with a β-sheet structure formed due to protein misfolding or denaturation. It is mainly deposited in the intercellular space, causing changes in the structure and function of tissues and organs, thus causing heterogeneous diseases with corresponding clinical manifestations. Studies in recent decades have shown that at least 20 kinds of amyloid are closely related to neurodegenerative diseases, diabetes and other chronic diseases, such as Alzheimer's disease, type II diabetes, Parkinson's disease and Huntington's disease. In addition, protein aggregates (some of which are amyloid) formed due to protein denaturation will bring great difficulties to the productio...
