The invention provides a configuration method of a cyclic prefix (CP), a base station and a terminal, wherein the method comprises the steps of respectively configuring the CP length of each subframe by the base station, dispatching the terminal in need of the first CP length at the subframe which is configured with the first CP length and dispatching the terminal in need of the second CP length at the subframe which is configured with the second CP length according to the transmission requirement of the terminal. The invention provides a method for flexibly configuring the CP length, not allthe uplink or downlink subframes are necessarily configured to extended CP by using the method for the terminal which supports the multipoint cooperation or relay transmission, however the subframe is respectively configured, part of the subframes are configured to extended CP, the terminal which supports the multipoint cooperation or relay transmission is dispatched on the subframe which is configured with the extended CP, and the terminal is dispatched on the subframe which is configured with the conventional CP by using the common transmission mode, thereby improving the transmission efficiency of the system and saving the resources of the system.
