When processing cellulosic biomass, it may be desirable for process efficiency purposes to operate the digestion unit without complete depressurization. The method of processing cellulosic biomass may comprise: providing a biomass conversion system comprising a pressurized zone and a digestion unit operably interconnected; providing cellulosic biomass at a first pressure; converting at least a portion of the cellulosic biomass to introducing the substance into a pressurized zone, and then pressurizing the pressurized zone to a second pressure higher than the first pressure; after pressurizing the pressurized zone, transferring at least a portion of the cellulosic biomass from the pressurized zone to a digestion unit, the a digestion unit at a third pressure less than or equal to the second pressure but greater than the first pressure; and digesting at least a portion of the cellulosic biomass in the digestion unit to produce a hydrolyzate comprising soluble carbohydrates in a liquid phase.
