Abstract

The invention discloses a method for parthenogenetic activation of buffalo oocytes after vitrification. According to the method, an optimized activation processing manner of combining ionomycin and 6-DMAP is adopted, and the conventional method for parthenogenetic activation of buffalo oocytes after vitrification is improved and optimized. Experiments prove that the parthenogenetic activation condition has a certain influence on the developmental potential of frozen-thawed buffalo oocytes. By applying the method for parthenogenetic activation of frozen-thawed buffalo oocytes, the activation rate can be improved so as to improve the survival rate and developmental potential of the frozen-thawed oocytes.

Description

technical field [0001] The invention belongs to the field of mammalian reproduction, and in particular relates to a parthenogenetic activation method after vitrification and freezing of buffalo oocytes. Background technique [0002] Buffalo is one of the livestock with the most development potential and development value considered by the Food and Agriculture Organization of the United Nations. Preserving eggs with genetic value by vitrification can improve the use value of buffalo eggs. At present, the vitrification process of buffalo oocytes is still very slow, and many researchers focus on the optimization of vitrification procedures, ignoring whether the activation conditions are also problematic. In addition to factors such as changes in ultrastructure after freezing and thawing, activation conditions are also an important factor in vitrification. [0003] Oocyte activation is a key technical link in microfertilization and nuclear transfer in livestock. Haploid and ho...
