[0022] (1) RNA extraction from carp

[0023] Take a female carp with a size of about 300 g, and after dissection, take mung bean-sized ovarian tissue (50-100 mg), add 1 ml of TRIzol reagent, and use a homogenizer for homogenization. The sample volume should not exceed the volume of TRIzol by 10℅.

[0024] Place the homogenized sample at room temperature (15-30° C.) for 5 minutes to completely separate the nucleic acid-protein complexes.

[0025] Centrifuge at 10,000 xg for 15 minutes at 4°C. The sample is divided into three layers: the bottom layer is a yellow organic phase, the upper layer is a colorless aqueous phase and an intermediate layer.

Abstract

The invention belongs to the technical field of aquatic animal genetic engineering, and particularly relates to an isolated antiviral natural immune protein TRIM32 (tripartite motif 32) for carps and antiviral activity. The nucleotide sequence of a gene of the antiviral natural immune protein TRIM32 for the carps is represented as 1-1986bp in SEQ ID NO: 1 and the coded amino acid sequence of the antiviral natural immune protein TRIM32 is represented as SEQ ID NO: 2. The test proves that expression of the cloned protein TRIM32 can influence proliferation of SVCV (spring viraemia of carp virus), and protein TRIM32 plays an important role in the life cycle of SVCV; the gene or the protein provides a new target for study of anti-SVCV medicine preparation.
