The method comprises the following steps of obtaining regional geographical information data, land utilization investigation data and an ecological protection red line evaluation preliminary result; pre-treating the obtained data; aggregating patches in the ecological protection red line evaluation preliminary result; extracting the land utilization investigation data intersected with the ecological protection red line evaluation preliminary result; merging patches of the same land class attribute in the extracted land utilization investigation data; dividing boundary patches of the land utilization investigation data, deducting a non-ecological protection red line land, and obtaining a preliminarily-corrected ecological protection red line; and based on the regional geographical information data and high-precision remote sensing images, further correcting the boundary of the ecological protection red line, and obtaining the finally-optimized regional ecological protection red line. The method improves the delineation accuracy of the conventional ecological protection red line.
