The detection method comprises the following steps: dissolving the capsaicin in the emplastrum by use of a methanol-tetrahydrofuran mixed solvent in volume ratio of 1:1, measuring the capsaicin content by use of the high-performance liquid chromatography, wherein the high-performance liquid chromatography condition is as follows: octadecylsilane chemically bonded silica is taken as a filling agent, and acetonitrile-0.1% phosphoric acid liquid in volume ratio of 46:54 is taken as a mobile phase, and the detection wavelength is 205nm. The detection method disclosed by the invention is strong in specificity and good in repeatability, the RSD of the sample parallel testing is only 1.58%; the linearity is good, the linearity relation with the peak area is good when the capsaicin is between 10microgram / ml to 100microgram / ml; the recovery rate is high, and the accuracy is high.
