Embodiment 1

[0011] The preparation of embodiment 1 rescue must be medicinal material extract

[0012] Get an appropriate amount of Bing medicinal materials, crush them, pass through a No. 1 sieve, add 8 times the amount of 70% (volume fraction) ethanol to the medicinal materials, heat and reflux for extraction twice, each time for 1 hour, filter, combine the extracts, put them in a rotary evaporator, Concentrate under reduced pressure at 80°C to a thick paste. Transfer the concentrated liquid to a decompression drying oven, continue drying and crushing at 80°C, and obtain the extract of the rescued Bing medicinal material.

Embodiment 2

[0014] 1. Experimental materials

[0015] 1.1 The drug under test: basic acid (extracted from basic acid, purity > 98%).

[0016] 1.2 Control drugs: lamivudine, oleanolic acid, extracts from the medicinal materials of Gyubing (obtained in Example 1)

[0017] 1.3 In vitro cell model: 2.2.15 cell line (HepG2.2.15 cells) transfected with hepatitis B virus. Inoculate HepG2.2.15 cells in DMEM medium (containing 10% fetal bovine serum, 380 μg / ml G418, 0.03% glutamine, 100 μg / ml each of penicillin and streptomycin) in 5% CO 2 , cultured in a 37°C incubator.

[0018] 2. Reagents and main instruments

[0019] Dimethyl sulfoxide (DMSO, AR), MTT (thiazolium blue, AR), hepatitis B virus surface antigen detection kit and hepatitis B virus e antigen detection kit were purchased from Shanghai Industrial Kehua Biotechnology Co., Ltd.

[0020] 3. Experimental method

[0021] HepG2.2.15 cells in the logarithmic growth phase were selected, digested with trypsin digestion solution, and dilut...

Abstract

The invention discloses application of rotundic acid in preparation of an anti-hepatitis B virus (HBV) medicine for the first time, and especially discloses application of the rotundic acid in preparation of medicines for inhibiting secretion of HBV surface antigen or HBV E antigen. In-vitro HepG22.2.15 cell pharmacological experiments prove that rotundic acid has excellent anti-HBV effect, and has the advantages of relatively low effective concentration and relatively small cytotoxicity. The rotundic acid has an effect for inhibiting HBsAg and HBeAg, has the inhibition rates for HBsAg and HBeAg respectively being 39/5 percent and 40.2 percent at 100mu g/ml, and has the inhibition effect for HBV superior to that of positive control lamivudine, oleanolic acid and ovate leaf holly bark medicinal material extract.

Description

technical field [0001] The present invention relates to a new application of basic acid, in particular to the use of basic acid in the preparation of anti-hepatitis B virus drugs. Background technique [0002] Hepatitis B caused by hepatitis B virus (Hepatitis B Virus, HBV) is a worldwide disease. According to statistics, the infection rate of hepatitis B virus in my country is about 60%-70%, and 300,000-500,000 people die of liver cirrhosis or liver cancer caused by HBV every year. Currently clinically used anti-HBV drugs mainly include interferon and nucleoside drugs. Although these two types of drugs have relatively good curative effects in the treatment of HBV infection, they all have the disadvantages of high price, many adverse reactions, easy drug resistance, and easy rebound after drug withdrawal. Therefore, it is very important to find new safe and effective anti-HBV drugs. [0003] Jubiying, also known as iron holly, bear bile wood, etc., is a plant of the famil...
