Embodiment

[0075] Materials and methods

[0076] Sows / gilts (gilts): Sow domestic pigs (8-12 months of age) will be used as recipients for embryo transfer and will be cared for as regular domestic pigs for the expected period of gestation and farrowing according to the licensed IACUC protocol And maintenance.

[0077] Estrus Synchronization and Insemination: Sows were given 6.8 mL of Matrix (tetraenestrone 2.2 mg / mL) mixed into the morning feed on days 11-22 of the estrus to synchronize estrus. Lutalyse (2cc) will be given on the last day of Matrix and after 4 days. Sows will be checked for heat twice a day starting on day 6 after the end of Matrix administration. After initiation is first detected, sows will be inseminated up to three times with semen from selected boars. Sows were checked for pregnancy between days 23-90 of gestation using Doppler ultrasound or transabdominal ultrasound with a linear 5 mHz transducer. Both forms of ultrasound are invasive and do not cause harm to t...

Abstract

Described herein is a method for producing a chimeric non-human animal expressing a human ETV2 gene comprising: a) generating an ETV2 null non-human animal cell, wherein both copies of the non-human ETV2 gene carry a mutation that prevents production of functional ETV2 protein in said non-human animal; b) creating an ETV2 null non-human blastocyst by somatic cell nuclear transfer comprising fusinga nucleus from said ETV2 null non-human animal cell of a) into an enucleated non-human oocyte and activating said oocyte to divide so as to form an ETV2 null non-human blastocyst; c) introducing human stem cells into the ETV2 null non-human blastocyst of b); and d) implanting said blastocyst from c) into a pseudopregnant surrogate non-human animal to generate a chimeric non-human animal expressing human ETV2.

Description

[0001] priority statement [0002] This application claims the benefit of priority to US Provisional Patent Application No. 62 / 127,330, filed March 3, 2015, which is hereby claimed, and which is hereby incorporated by reference in its entirety. [0003] sequence listing [0004] This document incorporates herein by reference the electronic sequence listing text file, which is submitted electronically via EFS-Web. The text file is called "1552048.txt," is 12,288 bytes long, and was created on March 1, 2016. Background technique [0005] Blood is the main organ of the human body. It consists of cells and plasma. The cellular portion includes red blood cells, white blood cells, and platelets. The plasma composition consists of water and dissolved compounds such as proteins, sugars or electrolytes, among others. [0006] Blood formation begins during gastrulation. The first hematopoietic lineage arose in the blood islands of the yolk sac. The differentiation of hematopoieti...
