Problems solved by technology

Using metabolic surfactant microorganisms to treat oily sludge can increase the solubility of petroleum hydrocarbons, which can improve the degradation efficiency of petroleum hydrocarbons, and use biological treatment to remove harmful substances such as PAHs. Biological treatment can simultaneously degrade aliphatic hydrocarbons and aromatics in oily sludge However, the biological treatment method has limitations such as being greatly affected by environmental factors and a long treatment period, so the treatment technology for biological treatment of oily sludge has not yet been put into large-scale application in China
At the same time, the current microbial degradation agents are mainly compound microbial agents. Various strains need to be cultured and activated separately and then mixed in a certain proportion. cumbersome operation
In the prior art, there is no technical solution for using a single bacterial strain to achieve relatively excellent degradation of petroleum hydrocarbons in oily sludge

Embodiment 1

[0058] Example 1: Degradation and repair test of oily sludge in Liaohe Oilfield

[0059] Oily sludge from Liaohe Oilfield was taken, air-dried and ground, passed through a 1mm sieve, and set aside. Pick a single colony in sterilized LB liquid medium, seal it with a gas-permeable sealing film, place the Erlenmeyer flask in a shaker at 120 rpm, and activate at 30°C for 24 hours. Weigh 40 g of the above-mentioned air-dried, ground and sieved oily sludge, add 10% inoculation amount of Pseudomonas stutzeri JN4 activated bacterial solution with OD600=1, adjust the water-soil ratio to 2:5 with distilled water, and seal it with a breathable sealing film. All Erlenmeyer flasks were placed in a constant temperature incubator at 30°C for static cultivation, samples were taken every 5 days, and the content of total petroleum hydrocarbons in the oily sludge was measured with an infrared spectrophotometer. The degradation curve is shown in the attached image 3 shown. It can be seen from ...

Embodiment 2

[0063] Example 2: Degradation effect of Pseudomonas stutzeri JN4 on petroleum hydrocarbons in oily sludge at different pH values

[0064] Oily sludge from Liaohe Oilfield was taken, air-dried, ground, and passed through a 1mm sieve. Weigh 40 g of the above oily sludge in a conical flask, and use dilute H 2 SO 4and NaOH to adjust the pH of the soil to 1, 3, 5, 7, 9, 11, respectively, according to the inoculation amount of 10%, add the activated bacterial liquid of Pseudomonas stutzeri JN4 with OD600=1, and adjust the water-soil ratio to 2:5 with distilled water, and use Breathable parafilm closure. All the Erlenmeyer flasks were placed in a constant temperature incubator at 30°C for static cultivation and samples were taken after 25 days, and the total petroleum hydrocarbon content in the oily sludge was determined by an infrared spectroscopic oil analyzer. The degradation effects of strain Pseudomonas stutzeri JN4 on petroleum hydrocarbons in oily sludge under different pH ...

[0065] Example 3: Degradation effect of Pseudomonas stutzeri JN4 on petroleum hydrocarbons in oily sludge at different temperatures

[0066] Oily sludge from Liaohe Oilfield was taken, air-dried, ground, and passed through a 1mm sieve. Weigh 40g of the above oily sludge into an Erlenmeyer flask, add 10% inoculum amount of Pseudomonas stutzeri JN4 activated bacterial solution with OD600=1, adjust the water-to-soil ratio to 2:5 with distilled water, and seal it with a breathable sealing film. The Erlenmeyer flasks were placed in constant temperature incubators at 10°C, 15°C, 20°C, 25°C, 30°C, 35°C, 40°C, 45°C, and 50°C for static culture, and samples were taken after 25 days. Spectrophotometer was used to determine the total petroleum hydrocarbon content in oily sludge. The degradation effects of strain Pseudomonas stutzeri JN4 on total petroleum hydrocarbons in oily sludge at different temperatures are as follows: Figure 5 shown. Depend on Figure 5 It can be seen that the...

Abstract

The invention discloses a highly effective degradation strain JN4 for petroleum hydrocarbons in oily sludge and application thereof, which belong to the technical field of bioremediation. The degradation strain JN4 is preserved in the China General Microbiological Culture Collection Center, the preservation number is CGMCC NO.14975, and the classification number is Pseudomonas stutzeri. The strainJN4 with a petroleum hydrocarbon degradation function which is disclosed by the invention is applied to treatment of oily sludge by degrading petroleum hydrocarbons. The degradation rate of the bacteria on total petroleum hydrocarbon (TPH) in oily sludge is 71.05 percent within 30 days. The single strain provided by the invention has obvious degradation capability on n-C10 to n-C30, and GC-MS (gas chromatography-mass spectrometry) analysis indicates that the strain has strong degradation capability on cycloalkane naphthane, indane and d-homoandrostane. The optimum degradation conditions of the bacteria on petroleum pollutants are 25 DEGC to 40 DEG C and the pH value of 5 to 11. The biological method is used in oily sludge remediation technology, and has the advantages of high effectiveness, low cost, environment-friendliness and the like.

Description

technical field [0001] The invention relates to the technical field of bioremediation, in particular to a bacterial strain for degrading and repairing oily sludge and its application in degrading oily sludge. Background technique [0002] Since the 20th century, petroleum has become one of the most important energy sources for human beings. While being exploited and used in large quantities, the harm caused by petroleum and its processed products to the environment and human beings has become increasingly serious. Since China's reform and opening up, my country's annual output of crude oil has exceeded 100 million tons. China has become one of the world's top ten oil-producing countries and has more than 400 oil fields and oil and gas fields for exploration and development, which are distributed in 25 countries accounting for about 3% of the country's land area. Provinces (municipalities) and autonomous regions. However, in the process of rapid economic and technological dev...
