Abstract

The invention relates to a microbiological integrity index-based wetland ecosystem health evaluation method. The method comprises the following steps: 1) selecting multiple wetlands which can completely reflect the overall water quality and the aquatic ecosystem condition in the to-be-evaluated area, adopting the multiple wetlands as sampling points, and measuring 13 indexes; 2) filtering water samples, shearing a filtered filter membrane, transferring the samples into a crushing tube for DNA extraction, and carrying out PCR (polymerase chain reaction) amplification on obtained DNA samples; distinguishing the samples throughdifferent barcode sequences, distributing the total sequencing reading to all the samples, removing fuzzy bases included in the sequences, removing sequences with the primer mismatching conditionand containing more than six basic groups at the same time, filtering out low-quality sequences, and removing a primer sequence; 3) applying the weighted average regressionmethod to obtainan optimal value of each classification unit, and then determining the sensitive or tolerance classification unit of each key environmental factor.
