[0048] Synthesis of fluorescent probes

[0049] Under argon protection, raw material a (0.7094g, 1mmol), 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide (EDC, 0.1917g, 1mmol), 1-hydroxybenzo Triazole (HOBT, 0.1351g, 1mmol), triethylamine (0.5ml), and dimethylformamide (DMF, 2ml) were dissolved in dichloromethane (10ml), and ice-bathed for 30min. Then raw material b (0.1531g, 1mmol) was added to it, overnight at room temperature; after the reaction was completed, the reaction product was separated and purified by thin layer chromatography (dichloromethane:methanol=15:1) for the next step of reaction .

[0050] Add stannous chloride (0.44g, 2mmol) to the product of the previous step (0.4228g, 0.5mmol), and dissolve in methanol (20ml), then add hydrochloric acid (3ml) dropwise, and heat and reflux at 75°C; Chromatography (dichloromethane / methanol=10 / 1) was monitored in real time to determine the end point of the reaction. The reaction is complete when the reactant sites disappear ...

Abstract

The invention relates to a two-photon fluorescent probe used for qualitatively detecting concentration of nitric oxide. The two-photon fluorescent probe is named TCO-TP, and includes coumarin and o-diaminobenzene structures, with two kinds of easily-obtained synthesized compounds being initial raw materials, the probe is prepared through two steps of simple and efficient synthesizing reactions, aftertreatment is easy, and purification is easy and convenient. The maximum absorption and the maximum emission of the probe are 348 nm and 440 nm respectively, and large Stokes shift (90 nm) is realized. During in-vitro detection of nitric oxide, the reaction sensitivity of the probe with nitric oxide is high, and the fluorescent response multiple is high. Reaction conditions of TCO-TP and nitricoxide are optimally sieved, and it is found that under physiological conditions (Ph 7.4 and 37 DEG C), the fluorescent response of TCO-TP and NO is optimal. The TCO-TP has quite good selectivity for nitric acid, and specificity detection can be conducted on nitric acid in complex cell and living body biological environments.

Description

technical field [0001] The invention belongs to the field of two-photon fluorescent probes, and in particular relates to a two-photon fluorescent probe for qualitative detection of nitric oxide concentration, a synthesis method and an application thereof. Background technique [0002] Nitric oxide (NO) is a small molecular free radical and an important signaling molecule in cells. It is widely distributed in various tissues in the living body, and plays a very important biological role in the regulation of heart and cerebrovascular, nerve regulation, and immune regulation. A large number of studies have shown that the release of an appropriate amount of NO can cause a series of normal physiological effects of the organism, while excessive or insufficient release of NO will produce a series of pathological effects, such as vascular endothelial dysfunction, neurodegenerative damage, cancer and inflammation, etc. Diseases endanger human health. [0003] Fluorescence imaging t...
