Embodiment 1

[0027] Example 1 Bacteriostat Screening and Culture Medium Preparation

[0028] Screening of bacteriostatic agents: ε-polylysine, isothiazolinone, streptomycin, and chlorothalonil were selected in advance by the Key Laboratory of Crop Cultivation and Germplasm Innovation of Nanjing Agricultural University and could be used for antibacterial culture of chrysanthemums. Bacteriostats, mixed in equal amounts 1:1:1:1 to prepare MS medium containing different concentrations of bacteriostats, the concentrations are 0.025g / L, 0.05g / L, 0.075g / L L, 0.1g / L, the prepared medium is divided into glass tissue culture bottles, 40ML per bottle, sterilized in a sterilizing pot at 121°C for 30min, cooled and solidified for later use. Select Penicillium griseoviridum, inoculate it in 1 / 4MS liquid medium, culture it on a shaker at 25°C and 20r / min until the OD value is 0.6-0.8, dilute it 10000 times to make a suspension (this concentration can ensure that There is the existence of Penicillium cin...

Embodiment 2

[0031] A tissue culture method for protecting the intellectual property rights of chrysanthemums, mainly aimed at tissue culture and propagation of existing tissue culture seedlings, using a special antibacterial medium and tissue culture method, specifically including the following steps:

[0032] 1) Medium formula

[0033] Antibacterial medium preparation: ε-polylysine 0.05g / L, isothiazolone 0.05g / L, streptomycin 0.05g / L, chlorothalonil 0.05g / L, sucrose 30g / L, agar 7g / L L, MS powder 4.74g / L, the pH of the medium adjusted by NaOH is 5.8-7.2. The prepared medium was divided into glass tissue culture bottles with a volume of 40mL / bottle, sterilized in a sterilizing pot at 121°C for 30min, cooled and solidified for later use.

[0034] 2) Propagation of chrysanthemum intellectual property varieties

[0035] Take the middle and upper young stems of the aseptic seedlings of chrysanthemum intellectual property varieties that need to be protected, inoculate them in ordinary medium ...

Abstract

The invention discloses a tissue culture method for protecting the intellectual property varieties of a chrysanthemum. The method comprises the steps of preparing the bacteriostatic culture medium ofmultiple kinds of bacteriostatic agent combinations and inoculating penicillium glaucum, which exists widely in the natural world and is easy to extract, in the bacteriostatic culture medium so as toenable the penicillium glaucum to be in a latent state under the action of a bacteriostatic agent, fungal contamination is not caused, the growing development condition of chrysanthemum tissue cultureseedlings cultured in the culture medium and the expression after hardening-seedling are both normal, and the yield and quality of chrysanthemum seedlings are both not influenced; the chrysanthemum tissue culture seedlings in the bacteriostatic culture medium are inoculated into a common culture medium, the tissue culture seedlings give rise to the fungal contamination within a wide range withoutthe inhibiting effect of the bacteriostatic agent, and the contamination rate is up to 100%. By the method, the intellectual property varieties of the chrysanthemum can be effectively protected whenthe production efficiency and quality of the chrysanthemum tissue culture seedlings are simultaneously guaranteed, only the units which own a variety granting right can obtain healthy tissue culture seedlings, and therefore, good varieties are prevented from illegal outflow.

Description

technical field [0001] The invention relates to the technical field of tissue culture, in particular to a tissue culture method for protecting intellectual property varieties of chrysanthemums. Background technique [0002] Chrysanthemum, also known as Jinying, Jiuhua, Juhua, Jiehua, and Nuhua, is a perennial herbaceous plant of the genus Chrysanthemum in the family Compositae. It is one of the top ten traditional famous flowers in my country and one of the four major cut flowers in the world. It has a long history of cultivation in my country. Historically, it is one of the most abundant ornamental plants. It has a variety of application forms and a wide range of uses, mainly for ornamental, edible, tea and medicinal purposes. In production, chrysanthemum is usually propagated by two methods of cutting and division, but for the large-scale seedling production of new varieties, especially for professional breeding units, the propagation rate of these two methods is far from ...
