Problems solved by technology

[0003] At present, the methods for determining glutathione reductase mainly include Elisa method and ultraviolet enzymatic method. The former requires manual operation, takes a long time, has cumbersome steps and high cost, while the latter has simple steps and low cost, and is widely used.
[0004] The existing kits for ultraviolet enzymatic glutathione reductase are mainly imported dry powder reagents, which need to be dissolved in buffer and distilled water before use. The dissolved reagent solution can only be stored at 2-8°C for two days. If you don't finish it will be a waste

Embodiment 1

[0042] A glutathione reductase detection kit, comprising the following components:

[0043] First reagent, including phosphate buffer, EDTA-NA2, ascorbate oxidase, potassium ferricyanide, preservatives and oxidized glutathione;

[0044] Secondary reagents, including tris buffer, EGTA, preservatives and reduced coenzyme.

[0045] The advantages of the present invention are:

[0046] (1) Both the first reagent and the second reagent are in liquid state, and can be stored at 2-8°C for 12 months after thermal stability test with thermal acceleration;

[0047] (2) Potassium ferricyanide in the first reagent can oxidize ferrous ions (Fe2+) in hemoglobin to ferric ions (Fe3+), and hemoglobin is converted into methemoglobin. Methemoglobin combines with cyanide ion (CN-) to generate stable cyanide hemoglobin, which has no effect on the detection of glutathione reductase;

[0048] (3) The use of EGTA as the second reagent can improve the linear relationship between the detection resu...

Embodiment 2

[0069] This embodiment is basically the same as Embodiment 1, the difference is:

[0070] In the second reagent, the concentration of EGTA is 4 mmol / L.

Embodiment 3

[0072] This embodiment is basically the same as Embodiment 1, the difference is:

[0073] In the second reagent, the concentration of EGTA is 6 mmol / L.

Abstract

The invention discloses a glutathione reductase detection reagent kit which is prepared from the following components: a first reagent and a second reagent. The first reagent is prepared from the components of a phosphate buffer solution, EDTA-NA2, ascorbic acid oxidase, potassium ferricyanide, a preservative and oxidized glutathione. The second reagent is prepared from the components of a trimethylamine methane buffer solution, EGTA, a preservative and reduced coenzyme. The first reagent and the second reagent are both liquid and can be stored for 12 months at 2-8 DEG C environment. Potassiumferricyanide can oxidize hemoglobin to produce stable hemiglobincyanide, and the influence of detection results is reduced. The second reagent adopts EGTA, the linear relationship between the detection results and the change rate of absorbance is can be improved, and adopting of two-point calibration is convenient. The invention further discloses application using the glutathione reductase detection reagent kit. The application comprises the steps that the first reagent and the second reagent are added into a sample solution separately and put into a biochemical analyzer for incubating, the absorbance is read and the change rate of absorbance delta A1 is calculated, and then the glutathione reductase activity of a sample is calculated.

Description

technical field [0001] The invention relates to the technical field of biochemical detection, in particular to a glutathione reductase detection kit and its application. Background technique [0002] Glutathione reductase (Glutathione reductase, GR) is a flavin enzyme, which can make the enzyme containing sulfhydryl group (-SH) in the reduced state and active state, maintain the integrity of red blood cells, and prevent hemoglobin oxidation. [0003] At present, the methods for determining glutathione reductase mainly include Elisa method and ultraviolet enzymatic method. The former requires manual operation, takes a long time, has cumbersome steps and high cost, while the latter has simple steps and low cost, and is widely used. . [0004] The existing kits for ultraviolet enzymatic glutathione reductase are mainly imported dry powder reagents, which need to be dissolved in buffer and distilled water before use. The dissolved reagent solution can only be stored at 2-8°C fo...
