Abstract

The invention discloses an application of sirtuin4 protein for improving mitochondrial functions and an action mechanism thereof, which belong to the technical field of biology. According to the invention, the rhodotorula glutinis sirtuin4 gene with the functions of reconstructing a mitochondrial network and improving the phenotype of mammalian senescent cells is recombined and expressed in mammalian cells. The experimental results show that the recombinant protein can activate related proteins of damaged mitochondrial autophagy; the SA-beta-gal activity level of mammalian cells is obviously reduced; the expression level of CK19 and beta1 integrin is enhanced; the sirtuin4 protein can be used for reducing the intracellular ROS level, obviously improving the intracellular ATP level and improving the mitochondrial morphology, so that the mitochondrial network is improved, the anti-aging capability of the cells is improved, and a new strategy is provided for treating mitochondrial dysfunction and preventing aging by using the sirtuin4 protein.

Description

technical field [0001] The invention belongs to the field of biological technology of strain protein application mechanism. More specifically, the present invention relates to the mechanism and application of a Rhodotorula viscosus sirtuin4 protein expressed by mammalian cells, which has the function of remodeling the mitochondrial network and improving the phenotype of aging mammalian cells. Background technique [0002] Evidence that Sirtuins genes in lower eukaryotes are related to lifespan provides a basis for mammalian Sirtuins genes to affect lifespan in mammals. Various experiments have proved that SIRT1 can be used for anti-aging. Recent studies have shown that brain-specific SIRT1 overexpression can prolong lifespan and delay aging in mice. Embryo development and postnatal life are severely impaired in inbred mice lacking SIRT1. A recent study confirmed that outbred mice lacking SIRT1 do not extend their lifespan through energy restriction. This suggests that en...
