specific Embodiment approach 1

[0042] Embodiment 1: In this embodiment, a YPB peptide segment has a sequence of P-P-R-K-K-K-R-K-H-R-L-W-A-A-H-C-R-K-I-Q-L-K-K-D-G-S-S.

specific Embodiment approach 2

[0043] Specific embodiment two: the application of the YPB peptide in this embodiment, the YPB peptide is synthesized with TAT and GGG, and then labeled with fluorescent FITC to obtain the FITC-TAT-GGG-YPB polypeptide; the FITC-TAT-GGG - The YPB polypeptide is used to suppress the combination of YY1 and EZH2, thereby inhibiting the proliferation of breast cancer cells and the growth of tumors.

specific Embodiment approach 3

[0044] Embodiment 3: The difference between this embodiment and Embodiment 2 is that the sequence of the FITC-TAT-GGG-YPB polypeptide is FITC-YGRKKRRQRRR-GGG-PPRKKKRKHRLWAAHCRKIQLKKDGSS.

[0045] Other steps are the same as in the second embodiment.

Abstract

The invention discloses a method and application for inhibiting breast cancer cell proliferation and tumor growth by using YPB peptide, and relates to the method and application for inhibiting breastcancer cell proliferation and tumor growth by using YPB peptide. The purpose of the present invention is to provide a substance that can inhibit the proliferation of breast cancer cells and tumor growth by inhibiting the binding of YY1 and EZH2. Through research on interaction between protein molecules, a binding region between two cancer-promoting proteins YY1 and EZH2 is identified, and according to the sequence of the binding region of YY1 on the EZH2 protein, a FITC-TAT-GGG-YPB polypeptide containing YPB is synthesized. The polypeptide can inhibit functions of YY1 and EZH2 by interfering binding of the two molecules, so as to inhibit the proliferation of breast cancer cells and the growth of transplanted tumors in nude mice. According to the invention, the method and application for inhibiting breast cancer cell proliferation and tumor growth by using YPB peptide can be obtained.

Description

technical field [0001] The invention relates to a method and application for inhibiting breast cancer cell proliferation and tumor growth by using YPB peptide. Background technique [0002] EZH2 (Enhancer of zeste homolog 2) is a histone methyltransferase that catalyzes the trimethylation modification of the 27th lysine (H3K27) on histone H3, which is an important symbol of gene silencing [Kim and Roberts 2016, Nat Med]. EZH2 is a member of the polycomb group (PcG) protein family, and its function is to repress gene expression by regulating chromatin remodeling. EZH2 is also an important functional component of polycomb repressive complex 2 (PRC2, repression complex 2), which is responsible for maintaining the repression of gene expression. PcG proteins are involved in epigenetic modification. Functionally, PcG proteins can be divided into two core protein complexes, PRC1 and PRC2, which can repress hundreds of genes, including a large number of transcriptional regulators...
