Abstract
The invention belongs to the technical field of biology, and particularly relates to a molecular marker for identifying Clupanodon thrissa and Konosirus punctatus based on a mitochondrial COI gene andapplication of the molecular marker. The Konosirus punctatus is widely distributed in coastal areas of China, and coastal areas of Korea and Japan. The Clupanodon thrissa is mainly distributed in east China sea, south China sea and coastal areas of Korea. Distribution areas of the two kinds of fishes are overlapped, the appearances of the two kinds of fishes are very similar, and the two kinds offishes are offshore economic fishes in China and are difficult to quickly identify from the appearances. A pair of specific primers (HB3) is designed and screened by comparing and analyzing specificloci of the COI gene in mtDNA. The specific primers HB3 can be amplified in the Clupanodon thrissa to obtain a single product of 537bp, and no product exists in the Konosirus punctatus. Two species can be distinguished by presence or absence of a PCR amplification product, and are verified in 30 randomly selected samples. The invention provides a method capable of quickly identifying the Clupanodon thrissa and the Konosirus punctatus without sequencing.
