Problems solved by technology

[0007] Current techniques still have several limitations: First, current techniques do not allow the distinction between contaminating RNA and RNA associated with the presence of replicating microbes—such as viruses—in the sample, contaminating RNA such as in the sample that is independent of viable microbes— - such as active virus particles - the presence of replication intermediates
In the case of positive-sense single-stranded RNA viruses, the small number of antisense RNA species produced hampers the sensitivity of existing detection techniques

Embodiment Construction

[0150] The present invention relates to a method for distinguishing live and dead microorganisms in a sample, preferably a cell sample. In particular, the method according to the invention is based on the distinction of transcriptionally active microbial nucleic acid sequences from inert microbial nucleic acid sequences in a sample, preferably a cell sample.

[0151] The method of the invention is particularly useful for distinguishing between (1) dead microorganisms - such as viruses, bacteria, archaea, fungi or protozoa - and inert microbial sequences; and (2) active (or transcriptionally active) and latent microorganisms - Examples include viruses, bacteria, archaea, fungi or protozoa.

[0152] Therefore, it can be appreciated that the present method is readily applicable to the detection of any kind of microorganisms, as well as the distinction between live and dead microorganisms.

[0153] In one embodiment, the microorganism is selected from viruses, bacteria, archaea, ...

Abstract

The present invention relates to a method for discriminating between live and dead microbes in a sample, by discriminating between transcriptionally-active and inert microbial nucleic acid sequences in the sample. In particular, the method according to the present invention is based on the comparison of levels of nucleotide substitution in a sample cultured in presence of an RNA-labelling agent. The present invention further relates to a diagnosis method of microbial infections in a subject; and to methods of assessing the risk of contamination of a sample, implementing the method for discriminating according to the present invention.

Description

[0001] field of invention [0002] The invention relates to a method for distinguishing living microorganisms and dead microorganisms in a sample by distinguishing the nucleic acid sequences of microorganisms with transcription activity and the nucleic acids of microorganisms with transcription inertness in the sample. In particular, the method according to the invention is based on the comparison of the levels of nucleotide substitutions in samples incubated in the presence of an RNA labeling agent. [0003] The invention also relates to a method of diagnosing a microbial infection in a subject; and a method of implementing the method for differentiation according to the invention to assess the risk of contamination of a sample. Background technique [0004] The ability to detect viruses in cells, and more generally, microorganisms, has many applications in the field of diagnostics, where it has been shown to be useful in identifying infectious agents that cause, for example,...
