The method comprises the following steps: (1) pretreatment of a detected object; (2) preparing 221 reference substance single standard stock solutions, and preparing a mixed standard working solution and a plurality of standard series solutions according to the concentration gradient; (4) establishing chromatographic separation conditions and mass spectrum conditions; establishing a first-level spectrum library; and establishing a secondary high-resolution mass spectrum database. The method has the beneficial effects that the ultra-high performance liquid chromatography-quadrupole-time-of-flight high-resolution mass spectrometry is used for establishing a method for detecting 221 illegally added chemical components in the health food, and a high-resolution mass spectrometry database of corresponding illegally added chemical substances is established. And compared with corresponding characteristic fragment ions in a high-resolution mass spectrum database, qualitative and quantitative risk screening and determination can be carried out on 221 illegally added chemical substances for health food supervision sampling inspection and risk monitoring.
