[0037] Take 12-hour fasting normal human plasma, quantitatively transfer it into a small beaker, adjust the serum density to 1.300g / ml with solid potassium bromide, and transfer it to 6 transparent centrifuge tubes after the solid potassium bromide is completely dissolved. Tube 10ml, and then carefully spread 28ml of sodium chloride solution with a density of 1.006g / ml along the tube wall with a long needle of a syringe, so as not to disturb the interface. After balancing, cover and seal, carefully put into the RPV-50T vertical rotor, put it into a Beckman optima L-100XP ultra-high-speed low-temperature centrifuge (Fullerton...

Abstract

The invention belongs to the technical field of biomedicine, in particular to a natural anti-oxidation low-density lipoprotein (oxLDL) IgM subclass antibody for suppressing atherosclerosis. The natural anti-oxidation low-density lipoprotein (oxLDL) IgM subclass antibody is characterized in that a Babl/c rat is bred with a high-cholesterol diet for 4 weeks under a condition of no special pathogen;splenic cells (B cells primarily) are separated, the B cells and myeloma cells are combined by a chemical method, and hybridoma cells are obtained; and oxidized low-density lipoprotein (oxLDL) is usedas an antigen, an indirect enzyme-linked immunosorbent assay (ELISA) experiment is carried out on the growth holes of the positive hybridoma cells, positive cloning holes are determined, and the required cloning positive cells are screened out by a limiting dilution method. The obtained cloning positive cells are cloned and multiplied, cells generating a monoclonal antibody are obtained, and thenatural oxLDL-resisting immunoglobulin M (IgM) subclass antibody 3A6 is obtained. The antibody can be used for lowering the formation of the atherosclerosis of the rat and provides a novel idea and anovel method for researching the generation, the development and the treatment of the atherosclerosis.

Description

technical field [0001] The invention belongs to the technical field of biomedicine, and in particular relates to a natural anti-oxidation low-density lipoprotein (oxLDL) immunoglobulin M (IgM) subclass antibody capable of inhibiting atherosclerosis. Background technique [0002] Atherosclerosis is a chronic disease of the vessel wall that can lead to myocardial infarction, heart failure, peripheral vascular disease and stroke. Multiple risk factors lead to the formation of atherosclerosis in different aspects. Endothelial dysfunction is the initial link of atherosclerosis, and various risk factors can cause endothelial damage. After the endothelium is damaged, plasma low-density lipoprotein (LDL) infiltrates into the arterial wall through the damaged endothelium, oxidizes into oxLDL, is phagocytized by macrophages, becomes foam cells, and gradually forms atherosclerotic plaques. Lead to plaque rupture, local platelet aggregation, blood vessel embolism, formation of myocardi...
