Ginkgo flavone tablet and preparation method thereof

A technology of ginkgo flavonoid tablets and ginkgo flavonoids is applied in the directions of medical raw materials derived from Ginkgo biloba, pill delivery, metabolic diseases, etc., which can solve the problem of the asynchronous absorption and in vivo release speed of hydrophilic flavonoids and lipophilic lactones, and the absorption and loss of ginkgo flavonoids. Slow release in the body, prolong the onset time of ginkgo flavonoids, etc., to achieve the effects of fast onset, fast absorption and in vivo release, and low tablet cost

Problems solved by technology

The current pharmacokinetic studies of the total flavonoids of Ginkgo biloba have shown that due to the difference in the ester-water partition coefficient between ginkgo flavonoids and ginkgolides, the absorption and release rates of hydrophilic flavonoids and lipophilic lactones are not synchronized in the body, and the lactones Class ingredients can affect the absorption process ...

Abstract

The invention discloses a ginkgo flavone tablet comprising a ginkgo flavone extract and an assistant, wherein the ginkgo flavone extract is a product obtained by extracting ginkgo leaves by using the method: crushing the ginkgo leaves, adding water, heating and extracting; cooling an extraction solution in the step (1) to below 15 DEG C and standing and filtering; drying filtrate in the step (2) and collecting a dried substance; dissolving the dried substance with an ethanol solution and standing; condensing supernate to obtain a concentrated solution; dissolving the concentrated solution with ethanol and adsorbing and purifying with macroporous resin; and washing until the color of effluent fluid becomes light and eluting with the ethanol solution to obtain the ginkgo flavone extract. The tablet can be quickly absorbed and released in body and has high bioavailability.

Example Embodiment

[0028] The following further describes the present invention in detail through specific embodiments to help understand the content of the present invention.
[0029] Ginkgo flavonoid tablets prescription:
[0030] Ginkgo flavonoid extract 250.0g
[0031] Pregelatinized starch 85-105g
[0032] Hypromellose 1.2-1.5g
[0033] Magnesium stearate 1.5-1.9g
[0034] Silica 25-40 g
[0035]
[0036] 1000 pieces
[0037] The preparation process is as follows:
[0038] 1. Extraction of Ginkgo Flavonoids:
[0039] (1) Take 100 kg of Ginkgo biloba leaves and coarsely crush them to 8 meshes, place them in a dynamic extraction tank, add ten times the amount of purified water and heat to 50±5°C, dynamically extract three times, and combine the extracts;
[0040] (2) Cool the extract to below 15°C, let it stand for 12 hours, and filter the extract to separate impurities and other substances;
[0041] (3) Then the filtrate is spray-dried, and the dried product is collected;
[0042] (4) Dissolve the dried product into ethanol solution, use 700l of 50% ethanol (v/v) solution, heat and reflux for extraction, concentrate, recover ethanol, and collect the concentrated solution for later use;
[0043] (5) The concentrated solution is adsorbed and purified with D140 macroporous resin to separate the ginkgo flavonoids, first wash with purified water until the effluent is light in color, and then use ethanol solution, usually 70% ethanol solution (v/v) to elute, to obtain ginkgo flavonoids In the extract, the average yield of ginkgo flavonoids was 3.30%. The content of total flavonoid glycosides was determined and analyzed by high performance liquid chromatography. The average content of total flavonoid glycosides in the product was 25.1% (w%), and lactones were not detected.
[0044] 2. Pass the ginkgo flavonoid extract through an 8 mesh sieve; pass the pregelatinized starch and magnesium stearate through an 80 mesh sieve respectively for use;
[0045] 3. Weigh hypromellose, add it to purified water or water for injection at 80-90℃, stir to dissolve it, place it until the temperature drops to room temperature, add 95% ethanol (v/v), stir evenly to obtain Hypromellose 2.0% (w%) and 50% ethanol (w%) solution as a binder;
[0046] 4. Weigh the prescription amount of Ginkgo brass extract and pregelatinized starch, and pour them into the mixing granulator according to the principle of equal increments. Mix for 5 minutes. The power of the cutting knife and the stirring knife is ≥50Hz. Add the prescribed amount of binder, the frequency of addition is 1 time/second, the mixing time is 90 seconds, and the mixing is uniform to prepare suitable wet granules.
[0047] 5. The material is then transferred to a one-step granulator for drying, and the drying temperature is not higher than 80°C. After the materials are dried, pour them into the feeder, add silica and magnesium stearate, select a 2mm size sizing screen, use a sizing machine to sizing to obtain dry particles. Sampling to determine the moisture content, the moisture content is controlled between 2% to 5% (w%). Pour the whole granules into a multi-directional motion mixer, add magnesium stearate according to the prescription, and mix for 10 minutes. Determine the content of intermediate particles, calculate the average tablet weight, and press the tablet with a Φ10mm shallow recessed die. The hardness range is 5-6N, and the friability is less than 1.0%.
