Embodiment 1

[0024] The preparation of embodiment 1 Bdellovibrio swimming body bacterial liquid

[0025] Inoculate 0.5mL 10 6 cfu / mL Aeromonas hydrophila (Aeromonas hydrophila, purchased from Guangdong Provincial Institute of Microbiology Culture Collection Center, number GIM1.172) in 100mL nutrient broth (10g peptone, 3g beef extract, 5g sodium chloride, 1000mL distilled water, pH 7.4) in liquid medium, cultured at 28°C for 20 hours, centrifuged at 6000rpm for 15 minutes, and precipitated with 5mL DNB (dilute nutrient broth) liquid medium (0.8g nutrient broth, 0.5g casein acid hydrolyzate, yeast extract Extract 0.1g, dissolved in 1000mL distilled water, pH value is 7.5) suspended, then added to 100mL DNB, inserted into 1mL 1×10 3 pfu / mL Bdellovibrio BDM01, cultivated at 28°C for 48 hours, centrifuged the culture solution at 7000rpm for 20min, centrifuged the supernatant of Bdellovibrio swimming body at 16000rpm for 20min, suspended the precipitate with 1mL DNB liquid medium, and obtained...

Embodiment 2

[0026] Application experiment of embodiment 2 Bdellovibrio swimming body in sea cucumber seedling cultivation

[0027] Implementation location: A sea cucumber farm in Shandong, breeding sea cucumber species: imitation sea cucumber, also known as gray cucumber

[0028] 1. Personal ginseng harvesting

[0029] Harvesting in natural sea areas; harvesting time: when the bottom water temperature of seawater reaches 15-16°C, ginseng will be harvested intensively 5 days before spawning; select individuals over 300g, over 3 years old and without damage.

[0030] 2. Ginseng storage

[0031] Stocking density 15 / m 3 , the water temperature for storage is 16-18°C, and the storage time is 7 days, and it is carried out in a storage pond. The water in the reservoir is finely filtered. Add Bdellovibrio swimming body bacterial liquid in water, make the concentration of Bdellovibrio swimming body in the cultivation pool reach 10 1 ~10 7 pfu / mL. During the short-term storage process, no ba...

Abstract

The invention discloses an application of bdellovibro swimmers in culturing young sea cucumbers, which is implemented by adding bdellovibro swimmer bacterial liquid to a water body and/or feeds. The implementation process specifically comprises the steps of preparing facilities and water quality conditions for culturing young sea cucumbers, catching parental sea cucumbers, rearing the parental sea cucumbers, hatching spawns and culturing larvae, and collecting and culturing young sea cucumbers. The key technology is as follows: adding the bdellovibro swimmer bacterial liquid with final concentration of 10-107pfu/mL to the water body and/or simultaneously soaking the feeds in the bdellovibro swimmer bacterial liquid with concentration of 10-107pfu/mL for 30 minutes. By applying the invention, the average survival rate of the cultured young sea cucumbers reaches more than 50%, and the water quality can be effectively improved. Experiment shows that the water can be kept unchanged in 20 days at most, thus saving the labor and cost and greatly improving the economic benefits. Besides, the survival rate of the cultured young sea cucumbers is high, the quality of the young sea cucumbers is good, and the invention is water-saving, energy-saving, environment-friendly and low-carbon.

Description

technical field [0001] The invention belongs to the field of biotechnology, and relates to the application of a bacterium, in particular to the application of the bdellovibrio swimming body bacterial liquid in sea cucumber seedling cultivation. Background technique [0002] Sea cucumber has been listed as one of the "Eight Treasures in the Sea" since ancient times, and has the reputation of "Ginseng in the Sea". According to research, sea cucumbers, especially northern spiny sea cucumbers, have the characteristics of high protein, low fat, and low sugar, and have high food value and medical value. In recent years, with the continuous improvement of the living standards of urban and rural residents and the enhancement of medical care awareness, the consumer demand for sea cucumbers has continued to rise, and the price has continued to rise, which has led to the rise of sea cucumber farming. Sea cucumber farming has become another new industry after shrimp farming. marine aqu...
