Problems solved by technology

[0005] Somatic embryos are mostly induced from suspension cultured cell lines and callus. In recent years, researchers have successfully induced somatic embryos in carrots, alfalfa, chicory, pine and cypress and other plants, but maize is the main genetic transformation recipient. material, so far there has been no report of successfully obtaining maize somatic embryos

Embodiment

[0052] 1. Material sources and selection of explants

[0053] The test material of the present invention is obtained by screening from 50 different corn inbred lines, put the immature embryos of 50 corn inbred lines into the callus induction medium, and put the callus induced for 20-30 days into In the subculture medium, the two materials of inbred line 1 and inbred line 2 appeared in a yellow, loose and granular embryogenic state. After being placed in the differentiation medium, they took root and sprouted at the same time without adding exogenous hormones. The differentiation rate is extremely high.

[0054] The two inbred lines were sown in the spring of 2008 and 2009, and immature embryos of the inbred lines were obtained under strict maize self-pollination conditions. Take the young ears of the above-mentioned inbred lines about 15 days after pollination in the ultra-clean workbench for alcohol spray disinfection, that is, peel off the bract leaves of the young ears, an...

Abstract

The invention provides a method for inducing somatic embryos of corn and belongs to the technical field of biological engineering. The method comprises the following steps of: inducing corn explants in a callus inducing culture medium; transferring the explants undergoing inducing culture to a subculture medium; propagating the somatic embryos in the subculture medium; and putting the propagated somatic embryos into differentiation culture medium for differentiation and seedling. By the method, a plurality of somatic embryos of corn can be induced with high efficiency, and the induced somaticembryos of corn have the characteristics of realizing the differentiation of both poles at the same time by long-time subculture infinite propagation and without the control of exogenous hormones, along with high differentiation rate and seedling rate; and because influence of exogenous hormones is avoided in the differentiation process, the mutation rate of somatic cells is relatively lowered atthe seedling stage of corn. The invention can provide an excellent and practical acceptor system for various trans-genetic methods (such as an agrobacterium-mediated method and a gene gun method).

Description

technical field [0001] The invention belongs to the technical field of bioengineering, in particular to a method for inducing corn somatic embryos. Background technique [0002] Corn is an important food crop, and the sown area reached more than 29 million hectares in 2010 2 , corn not only occupies an important position in my country's agricultural production, but also is more and more widely used in industrial production. [0003] In maize genetic transformation, the recipient transformation system with embryogenic callus culture and regeneration system is the preferred method. The explants of maize receptor system to induce callus generally choose various tissue parts with low degree of cell differentiation and strong meristematic ability, such as: pollen, anther, young ear, immature embryo, cotyledon node, hypocotyl, stem node and Bud tip and so on. Green and Phillips (1975) reported for the first time that callus was induced from immature maize embryos and successful...
