Embodiment 1

[0080] Example 1. Salt tolerance-related marker screening

[0081] 1. Screening of a single marker

[0082] First, 4 materials with different salt tolerance (2 typical salt-tolerant materials Zhong 9835 and Zhong 07, 2 typical salt-sensitive materials Zhong S9612 and Xinyan 96-48) were selected for SSR primer screening ( figure 1 ). 246 pairs of polymorphic primers were initially screened out of 5053 pairs of SSR primers, accounting for 6.21%. These 246 pairs of primers were used as candidate primers for further research.

[0083] Using 6 materials: 9835, Zhong 07, Zhong 9806, Zhong S9612, Xinyan 96-48 and 16 pairs of Lumianyan, 246 pairs of SSR were screened for further re-screening, and 95 pairs with obvious differences, clear and easy to identify band patterns were selected The primers were used for PCR amplification of 48 test materials. Through careful observation and analysis of the electrophoretic bands, it was found that 5 markers (CSHES149, HAU773, HAU1058, HAU191...

Embodiment 2

[0133] Embodiment 2, carrying out salt-tolerant cotton identification with marker combination

[0134] The details of the 11 materials used in this example are as follows: J1: Zhongjian 9901; J2: Zhongjian 9902; J3: Zhongjian 9903; J4: Zhongjian 9904; J5: Zhongjian 9905; J6: Zhongjian 9906; J7: Zhongjian 9907; J8: Zhongjian 9908; J9: Zhongjian 9909; J10: Zhongjian 9910; J11: Zhongjian 9911;

[0135] The public can purchase the above 11 materials from the National Cotton Germplasm Bank of the Cotton Research Institute of the Chinese Academy of Agricultural Sciences.

[0136] (1) Single plant detection method

[0137] 1. Materials: 11 materials (J1~J11).

[0138] 2. Three-marker combination: HAU1918(T4), HAU1058(T3A and T3B) and TMB1288(S1)

[0139] 3. Method:

[0140] The DNA of one individual plant from each material was randomly selected, and then used as a template for SSR-PCR amplification and detection of amplified products.

[0141] According to the individual plant ...

Abstract

The invention discloses a method and special SSR (Simple Sequence Repeat) markers for identifying salt-tolerant cotton. The primer compositions for auxiliarily identifying salt-tolerant cotton are HAU1918, HAU1058 and TMB1288. The experiment proves that the method disclosed by the invention has stable and reliable result, is simple to operate and is economic and efficient. Compared with the morphological identification method, the method disclosed by the invention has the advantages of high efficiency, high accuracy, time saving, labor saving and the like, is free from the restrictions of seasons, has wide application prospects, and can be used for identifying and evaluating salt tolerance of cotton in production practice. Therefore, the SSR marker compositions and method for identifying salt-tolerant cotton can have wide application prospects in the field of salt tolerance identification.

Description

technical field [0001] The invention relates to a method for identifying salt-tolerant cotton and a special SSR marker. Background technique [0002] The salt tolerance of cotton is a very complex trait, involving a variety of physiological and biochemical metabolic pathways and many gene regulation. There are large differences in salt tolerance between varieties, different growth stages of the same variety, and different tissues and organs. There may be many different mechanisms of salt tolerance. So far, no unified understanding has been formed, and there are still different opinions in many fields and argue. The complexity of salt tolerance of cotton brings great difficulties to the selection and identification of salt-tolerant varieties. If several markers related to cotton salt tolerance can be screened, and the salt tolerance of cotton can be directly identified and evaluated at the molecular level, it will be a major breakthrough in the identification method of cott...
