Embodiment 1

[0033] Example 1: Animal experiment of different doses of Artemisia polysaccharides on mouse immune model antigen ovalbumin (OVA)

Embodiment 1

[0034] The preparation method of present embodiment one artemisia polysaccharide is:

[0035] Step 1: Degrease fresh or dried Artemisia argyi powder with petroleum ether, add distilled water for ultrasonication, add absolute ethanol to stir and precipitate the filtrate after concentration, centrifuge the precipitate, and dry it in vacuum to obtain a crude product of Artemisia argyi crude polysaccharide;

[0036] Step 2: Use Sevage reagent (chloroform: n-butanol ratio: 4:1) to remove protein, centrifuge, combine supernatant, repeat the above operation several times, collect supernatant, add ethanol, let stand overnight, and precipitate Centrifuge and dry in vacuum to obtain the crude polysaccharide of Artemisia argyi.

Embodiment approach

[0038] Experimental material: Artemisia argyi polysaccharide mother liquor configuration: 0.1g of Artemisia argyi crude polysaccharide was dissolved in 10ml of normal saline to prepare a mother liquor of 10mg / ml. Prepare OVA mother solution: dissolve 0.01g OVA in 10ml of normal saline, and prepare a 1mg / ml mother solution.

[0039] Animal immunization: 6-8 week-old ICR mice were randomly divided into 8 groups, 6 mice in each group, and the experimental groups are shown in Table 1.

[0040] Table 1 Immune grouping of mice

[0041]

[0042] Each group of vaccines was dissolved in normal saline, and each mouse was injected with 100 microliters, subcutaneously on day 0, and re-injected at an interval of 14 days after the first injection, a total of two injections.

[0043] In order to detect the level of humoral immunity in mice, blood was collected after subcutaneous injection, the serum of mice was separated, and IgG and IgG were detected by indirect ELISA respectively. 1 a...

Abstract

The invention provides application of alpine yarrow herb polysaccharide as a vaccine adjuvant. Through an animal grouping and immune method, ELISA testing of titer and cellular immunological activity of an anti-OVA specific antibody in mouse sera and other experiments, results prove that the alpine yarrow herb polysaccharide can serve as an adjuvant for protein vaccines or polypeptide vaccines or inactivated vaccines or attenuated live vaccines or nucleic acid vaccines, and the immunological enhancement effect of the alpine yarrow herb polysaccharide is obviously better than that of an aluminum adjuvant. The alpine yarrow herb polysaccharide has the advantages of being small in side effect, wide in source and easy to produce. Moreover, the preparation process of the alpine yarrow herb polysaccharide is simple, cost is low, yield is high, and large-scale production is facilitated.

Description

technical field [0001] The invention relates to an immune adjuvant, in particular to the application of Artemisia polysaccharides in vaccine adjuvants, and belongs to the field of biotechnology. Background technique [0002] Artemisiarupestris L is a perennial herb of the genus Artemisia in the Asteraceae family, mainly distributed in the Altai Mountains, Tianshan Mountains and Kunlun Mountains in Xinjiang, my country. Artemisia argyi is a commonly used medicine in Xinjiang Uighurs. It is used to treat hot colds, fever, headaches, hepatitis, and urticaria. The compound preparation with Artemisia argyi as the main drug has been marketed in Xinjiang for viral colds and allergic rhinitis, and has been widely used clinically. In the past ten years, researchers have carried out in-depth and meticulous research on its chemical composition and pharmacological effects, and found that Artemisia argyi contains flavonoids, polysaccharides and other chemical components, which have anti...
