Embodiment 1

[0040] Example 1: Determination of absolute concentration of piRNA in seminal plasma by TaqMan probe real-time fluorescent quantitative PCR method

[0041] (1) The subjects of the study were infertile persons who did not take any contraceptive measures within 2 years after marriage. Age-matched males who had given birth for less than 2 years were used as normal controls. Semen was collected from all subjects after 3 to 5 days of abstinence. WLJY -9000 Weili color sperm quality detection system (Beijing Weili Company) for sperm quality and function analysis. The analytical standards were all carried out according to the World Health Organization (WHO Laboratory Manual for Human Semen Examination and Processing, 5th edition). After the semen analysis, it was centrifuged at 1000g for 10min to collect the supernatant, and then centrifuged at 12000g for 5min to collect the supernatant seminal plasma.

[0042] (2) A total of 20 cases of asthenic infertility were collected (no histo...

Embodiment 2

[0046] Example 2: The RiskScore risk coefficient scoring system calculates and estimates each sample

[0047] First, draw the ROC curve for the data of the primary screening group, and calculate the RiskScore and Cut-off value through the mathematical analysis process, and the calculated Cut-off value is actually 3.57 (see Table 2).

[0048] Then use the following formula to score patient i and evaluate its risk coefficient, RiskScore i A score higher than the Cut-off value is a high risk factor, representing patient i with weak sperm motility and reproductive dysfunction. Scores below the cut-off value represent normal sperm motility.

[0049] RiskScore i =∑ n j=1 W j ·S ij

[0050] In the above formula, "s" of each piRNA is based on the lower limit of 5% of the normal control group as a reference value, that is, it is recorded as 1 if it is lower than the lower limit of 5% of normal, and 0 otherwise. "s ij " is the value obtained by piRNAj relative to samplei, and "...

Abstract

The invention discloses a seminal plasma piRNA marker combination reflecting male sperm activity and judging reproductive dysfunction and application thereof. Four piRNA markers include DQ570956, DQ571813, DQ575659 and DQ575661. The combination can be used for detecting sperm activity and predicting reproductive dysfunction caused by low sperm activity. Seminal plasma is easy to obtain, other tissue is not needed, and noninvasive examination is realized; seminal plasma piRNA can reflect pathological and physiological conditions of the whole spermatogenesis stage, detection accuracy can be improved by taking the seminal plasma piRNA as an auxiliary detection index, the pathological and physiological conditions in the whole spermatogenesis stage are reflected on the molecular level, and a potential target is provided for diagnosis and treatment of male reproductive dysfunction; a numerical value is calculated for each testee by a danger coefficient mathematical calculation method, and sperm activity can be reflected directly and visually through the numerical values, so that interference caused by subjective factors and lack of experience is avoided.

Description

technical field [0001] The invention belongs to the field of biotechnology, and relates to seminal plasma piRNA markers reflecting male sperm motility or combinations and applications thereof. Background technique [0002] Infertility has become a reproductive health problem worldwide. According to incomplete statistics, there are currently about 186 million people in the world suffering from infertility problems, of which male infertility accounts for about half, and it has been on the rise in recent years (1). Nearly 75% of male infertility patients are diagnosed as congenital idiopathic, yet the molecular mechanisms responsible for male infertility remain unclear (2). At present, clinical reproductive laboratory inspection items generally include biochemical inspection of semen, acid phosphatase, lactate dehydrogenase-X and other enzyme inspections, and semen immunological inspection, but these technologies have certain defects and cannot directly reflect testicular sper...
