The method is characterized by comprising the following steps: preparing hydrogen-rich water, controlling hydrogen addition amount and preparation time, discontinuously operating and preparing a series of standard samples from low hydrogen content to high hydrogen content; dividing each standard sample into two parts, and measuring a redox potential value of one standard sample by a redox potentiometer; carrying out titration by a titrant, calculating actual hydrogen concentration, repeating the above process to obtain correspondence between hydrogen concentration of hydrogen-rich water and the redox potential value, and drawing a curve chart of the correspondence between the hydrogen concentration of hydrogen-rich water and the redox potential value. A potential value of hydrogen-rich water with unknown hydrogen concentration can be measured by the use of a redox potentiometer, so as to calibrate concentration of hydrogen in water. The method of the invention is a calibration method which has advantages of simple equipment, convenient operation, reliable data and low cost. The method can be widely applied in calibration of hydrogen concentration of hydrogen-rich water.
