Embodiment 1

[0031] 1. Extraction of Genomic DNA from A. tomentosa

[0032] (1) Material

[0033] Salt-tolerant velvet wax H (Fraxinus velutina Torr) and salt-sensitive velvet wax S. Take the fresh leaves that have just grown, seal them with a fresh-keeping bag, put them in an ice box, and bring them back to the Shandong Academy of Forestry Sciences, freeze them in liquid nitrogen, and store them in a -70°C refrigerator for later use.

[0034] (2) Using the CTAB method to extract genomic DNA from leaves of A. tomentosa

[0035] 1) Preheat the prepared CTAB extraction buffer solution in a water bath at 65°C for 30 minutes, and place chloroform:isoamyl alcohol (24:1), absolute ethanol, and 70% ethanol in a -20°C refrigerator for pre-cooling. The mortar needs to be pre-cooled in advance, and then pre-cooled with a small amount of liquid nitrogen before use.

[0036] 2) Take 0.1-0.2g of white wax young leaves, add sufficient liquid nitrogen (add in two times) and grind to powder, quickly tr...

Abstract

The invention discloses an SSR molecular marker for the salt tolerance of Fraxinus velutina. Primers of loci of the SSR molecular marker for the salt tolerance of Fraxinus velutina include the primer S78 and the primer S95. The SSR molecular marker is expected to become a standard primer for identifying the salt tolerance of Fraxinus, and the accurate molecular marker is provided for rapid identification of salt-tolerant germplasm of Fraxinus and salt-tolerant breeding of Fraxinus. By means of the SSR molecular marker, the purpose of identifying salt-tolerant Fraxinus and salt-sensitive Fraxinus from the perspective of molecules is achieved, and a reference and a basis are provided for research such as salt tolerance molecular identification of Fraxinus.

Description

technical field [0001] The invention relates to a molecular marker, in particular to a salt-tolerant SSR molecular marker of white wax tomentosa, belonging to the technical field of molecular biology. Background technique [0002] Salt damage is a common abiotic stress for plants in the Yellow River Delta region, and it is one of the main environmental factors affecting plant growth and yield. Therefore, the breeding of new salt-tolerant plant varieties has always been the focus of research at home and abroad. Salt tolerance of plants is a complex quantitative trait involving the coordination of many genes and mechanisms of salt tolerance (Hao Derong et al., 2013). It is very difficult to identify salt-tolerant and salt-sensitive materials by morphological methods, especially to screen salt-tolerant individuals from hybrid progenies of salt-tolerant and salt-sensitive plants, which usually requires physiological assays and salt-tolerance tests, which are time-consuming, lab...
