The semisolid medium consists of the following components in percentage by weight: 2.46-2.48% of a CD FortiCHO medium and 2-3% of methyl cellulose. The CHO-S cell semisolid medium is prepared by the following steps: fully uniformly mixing the 2*CD FortiCHO and the 2*methyl cellulose according to a volume ratio of 1 to 1, conducting centrifuging so as to remove particles which are invisible to naked eyes, and collecting upper centrifugal liquid. The CHO-S cell semisolid medium disclosed by the invention can be used for replacing an imported CHO-S cell semisolid medium in cultivating CHO-S cells; cells are great in growth, high in protein expression quantity and good in growth status; and research and development cost and production cost can be greatly reduced.
