Embodiment 1
[0048] Expression of β-catenin in Glioma Patients
[0049] Previous studies have shown that AQP1 and β-catenin in glioma cells are closely related. On this basis, we hypothesized that this association would also apply to in vivo studies. Previous studies have shown that β-catenin is overexpressed in glioma, and its high expression is related to the malignancy of glioma (Liu et al.2011, Rossi et al.2011). In our study, β-catenin protein was mainly expressed in different intensities and locations in the cytoplasm ( Figure 4 D). Compared with low-grade (grade 1 and 2) gliomas, high-grade (grade 3 and 4) gliomas have a large amount of intracytoplasmic β-catenin protein expression. However, no significant nuclear staining was observed in specimens, including high-grade gliomas. High expression of β-catenin protein in grade 1 glioma and 39% of grade 2 glioma (n=24), but β-catenin in 55% of grade 3 glioma (n=30) and 58% of GBM (n=38) High expression of -catenin protein. For the...
Embodiment 2
[0051] Relationship between β-catenin protein expression and prognosis in patients with glioma
[0052] Kaplan-Meier analysis showed that high expression of β-catenin protein was associated with OS (P=0.034, Figure 5 E) and PFS (P=0.005, Figure 5 F) Significantly correlated. The median overall survival rate of patients with high expression of β-catenin protein was 31 months (95% Cl: confidence interval, 0.00-89.77), while the overall survival rate of patients with low expression of β-catenin protein (comprehensive score: 0-2) was 27 months (95% CI: confidence interval, 19.33-34.67). The median PFS of patients with high expression and low expression were 18 (95% Cl: confidence interval, 12.18-23.82) and 22 months (95% Cl: confidence interval, 7.65-36.36), respectively.
Embodiment 3
[0054] Relationship between AQP1 and β-catenin protein in glioma patients
[0055] Next, we investigated the relationship between AQP1 and β-catenin protein in glioma patients. Increased amounts of AQP1 and β-catenin proteins corresponded to histological grades. In addition, AQP1 labeling index was positively correlated with β-catenin protein expression (rs=0.168, P=0.022). High expression of AQP1 was significantly correlated with high-grade glioma and high expression of β-catenin protein.
