Embodiment 1

[0033] refer to figure 1 , a double-layer membrane for spinal cord therapy, comprising a dense outer layer 1 and an inner layer 2 from top to bottom, the dense outer layer 1 is a polylactic acid membrane, and the inner layer 2 is a loaded seed cell and mesoporous biological activity A chitosan film of a glass sphere, the mesoporous bioactive glass sphere loaded with bioactive factors. The bioactive factor is nerve growth stimulating factor. The periphery of the double-layer film is sealed with medical bio-glue. The seed cells are umbilical cord-derived mesenchymal stem cells, namely HUMSCs. The dense outer layer 1 is used to prevent the active factors and stem cells of the inner layer 2 from penetrating in this direction and seeping out to the outside through this layer of membrane. The purpose of providing stem cells and active factors.

Embodiment 2

[0035] Compared with Example 1, the difference of Example 2 is that the mesoporous bioactive glass spheres are replaced by ethylene glycol lactic acid copolymer (PEG-PLA) nano microspheres.

Abstract

The invention provides a double-layer membrane for spinal cord treatment. The double-layer membrane comprises a dense outer layer and an inner layer from top to bottom, wherein the dense outer layer is a polylactic acid membrane; the inner layer is a chitosan membrane supported with seed cells and mesoporous bioactive glass spheres; and bioactive factors are supported in the mesoporous bioactive glass spheres. The double-layer membrane is square or circular. The bioactive factors refer to nerve growth stimulating factors. The double-layer membrane aims to solve the problems in the prior art that a single damaged (but not dead) nerve tract cannot be repaired and treated; the problem that targeted release of the medicine cannot be realized by the conventional drug delivery system, and high concentrations of damaged local factors cannot be guaranteed, the problems that most of drug delivery systems are easy to degrade and low in drug-loading rate and drug release cannot always penetrate through the spinal cord injury repair process, and the problems that the drug delivery system is extremely large in volume, possibly compresses the spinal cord and is unfavorable for suture of duramater spinalis so as to cause leakage of a cerebrospinal fluid.

Description

technical field [0001] The invention relates to a double-layer film for treating spinal cord. Background technique [0002] Currently, the most common route of administration of nerve growth-promoting or nerve regenerative drugs is intravenous or intrathecal injection. There are also gel-binding active factor patches applied on the surface of the spinal cord, and L-polylysine-binding active factor as a scaffold implanted in the spinal cord to solve the problem of factors passing through the blood-brain barrier. [0003] Both drug delivery systems and stem cell bioscaffolds can be used to treat spinal cord injuries, but the combination of the two has not been seen so far. A similar combination has been used in the method and material of stem cell three-dimensional scaffold for bridging nerves, such as (a composite repair material and its scaffold for bridging defective nerves CN201510716279.8). However, the current treatment methods for bridging nerves with three-dimensio...
