Embodiment approach 1

[0064] The invention relates to a microfluidic device, comprising:

[0065] a cavity having microstructures disposed on its inner wall, the microstructures being configured such that when the solution is injected into the cavity, the solution forms bubbles at the microstructures; and,

[0066] The body wave generating device is used to generate body waves to resonate the above-mentioned bubbles to generate a flow field.

[0067] In one embodiment, the microfluidic device further includes a surface acoustic wave generating device for generating surface acoustic waves and controlling the position of particles in the solution.

[0068] An embodiment of the present invention provides a microfluidic device, such as figure 1 As shown, the device includes a cavity 110, a bulk wave generating device 120 and a surface acoustic wave generating device 130;

[0069] The interior of the cavity 110 is provided with microstructures, and when the solution is injected into the cavity 110, th...

Embodiment approach 2

[0110] Embodiments of the present invention provide a system for introducing substances into cells, such as Figure 5 As shown, the system includes an injection device 510 and the microfluidic device 520 provided in Embodiment 1 above;

[0111] The injection device 510 is used to inject a solution containing cells into the cavity of the microfluidic device 520, and the injected solution generates bubbles at the microstructure of the cavity;

[0112] The above-mentioned microfluidic device 520 is used to generate body waves, make the above-mentioned bubbles resonate, cause the vibration of the solution in the cavity, generate a flow field in the cavity, and each position in the flow field corresponds to a different shear stress, and control the cells in the cavity. The position of the flow field relative to the microstructure makes the cells flow through the preset position when flowing in the flow field, and the cells generate reversible perforation under the action of the she...

Embodiment approach 3

[0127] An embodiment of the present invention provides a method for introducing substances into cells. This method applies the system for introducing substances into cells in the above-mentioned embodiment 2. In this method, the body wave generation device in the microfluidic device can precisely control the flow of cells. The position in the field relative to the microstructure makes each cell pass a preset position when flowing in the flow field, thereby precisely controlling the shear stress on the cells.

[0128] Among them, the substances introduced into the cells in the embodiment of the present invention can be gene fragments, drug molecules, etc., and the user can put the substances to be introduced into the cells into the solution according to actual needs, and then use the method provided in the embodiments of the present invention to introduce the substances into the cells.

[0129] Using the method provided by the embodiments of the present invention to introduce su...

Abstract

The invention provides a microfluidic device and a system and a method for introducing a substance into a cell. The microfluidic device comprises a cavity channel, a body wave generating device and asound surface wave generating device, wherein a microstructure is arranged on the inner wall of the cavity channel and is configured in a manner that a solution can forms bubbles at the microstructurewhen being injected into the cavity channel; the body wave generating device is used for generating body waves to drive the bubbles to resonate so as to generate a flow field; and the sound surface wave generating device is used for generating sound surface waves so as to control the positions of particles in the solution. According to the microfluidic device, the positions of the particles in the solution can be precisely controlled by virtue of the sound surface wave generating device, and therefore, the shear stress borne by the particles in the solution can be precisely controlled.

Description

technical field [0001] The invention relates to the field of biotechnology, in particular to a microfluidic device, a system method and a method for introducing substances into cells. Background technique [0002] With the introduction of gene therapy theory and continuous in-depth research, human beings have entered a new stage of development in the study of thoroughly overcoming malignant tumors, hereditary diseases, and infectious diseases. The method is to introduce normal genes or genes with therapeutic effects into human target cells to correct gene defects or functional disorders, so as to achieve the purpose of treating diseases. How to efficiently and safely deliver exogenous substances from extracellular to intracellular is the key to gene therapy. [0003] At present, the methods for introducing substances into cells include viral gene introduction, injection, gene gun method, electroporation and sonoporation. The cells are shaken to make the bubbles evenly scat...
