The method is divided into two parts of initial value calibration and optimization calibration, wherein the initial value calibration stage is of controlling the three-coordinate measuring machine to respectively move for M1, M2and M3 times along three axes of X, Y and Z so as to measure M1 + M2 + M3 + 1 points including an initial position in a standard ball, and calculating the initial estimation direction of the laser beam and the initial estimation position of the center of the standard ball in the coordinate system of the three-coordinate measuring machine; and the optimization calibration stage is of controlling the three-coordinate measuring machine to scan and measure a large number of point clouds on the surface of the standard ball, and using an optimization method to optimize the laser beam estimation direction obtained at the initial value calibration stage so as to obtain an accurate direction of the laser beam in the measurement coordinate system of the three-coordinate measuring machine.
