Embodiment 1

[0036] The present embodiment constructs a kind of laryngeal precancerous lesion animal model, the method is as follows:

[0037] First, measure the distance from the Wistar rat incisors to the bilateral vocal cords and the root of the epiglottis, and determine the throat depth of oral smearing, with an average of about 12mm. The operation process is as follows: figure 1 A and B are shown.

[0038]Then, 36 10-week-old male Wistar rats were equally divided into two groups according to body weight, namely the control group and the model group, with 18 rats in each group. The laryngeal mucosa of Wistar rats in the model group was smeared with 4NQO dichloromethane mixed solution with a volume fraction of 1%, once every two days, and each rat was smeared for 1.5 minutes each time, for a total of 20 weeks. The Wistar rats in the control group were smeared with physiological saline on the throat mucosa, once every two days, and the smearing time was 1.5 minutes per rat, for a total ...

Abstract

The invention relates to a laryngeal precancerous lesion animal model and a construction method and application thereof. The construction method comprises the steps that a dichloromethane solution dissolved with 4-nitro quinoline-1-oxide is smeared on the laryngeal mucosa of an animal, and the laryngeal precancerous lesion animal model is obtained. 4NQO and dichloromethane are combined to achievea synergistic carcinogenic effect, so that the time required for successfully making the model by single drugs can be obviously shortened; the model is similar to a human patient, the laryngeal mucosaof the model turns white, spinous protrusions appear on the surface, or plaques and superficial ulcer appear on the surface of the laryngeal mucosa, and the model is highly similar to the patient inperformance and has good clinical symptom simulation; secondly, the 4NQO can be directly dissolved in the dichloromethane, so that the solubility and the stability of the 4NQO are improved; and a prepared mixed solution has certain viscosity and can be in better contact with and attached to the laryngeal mucosa when smeared on the laryngeal mucosa, and the stimulation time of cancerogen to the laryngeal mucosa is obviously prolonged. The construction method does not induce gastrointestinal tumors and reduces interference factors.

Description

technical field [0001] The invention belongs to the field of biotechnology, and in particular relates to an animal model of laryngeal precancerous lesions and its construction method and application, in particular to an animal model which well simulates human laryngeal precancerous lesions and its construction method and application. Background technique [0002] The occurrence, development, invasion and metastasis of cancer is a very complex process, and so is laryngeal cancer, which involves the participation of multiple genes and factors. The result of the interaction of various factors such as inactivation. Early detection of laryngeal cancer is especially critical in the treatment of laryngeal tumors. We need to understand the pathogenesis of laryngeal cancer and intervene early on the precancerous lesions, which is also one of the important treatment methods. In the process of laryngeal precancerous lesions to laryngeal cancer, different precancerous lesions have diff...
