Use of succinic acid as a biomarker for cancer diagnosis and treatment

The present disclosure demonstrates that cancer cells secrete succinate into the extracellular environment, which in turn increases the migration ability of macrophages and mediates cancer-associated macrophage polarization. In mouse models, succinate not only induces cancer cell epithelial-mesenchymal transition, enhances cancer cell migration ability, but also promotes cancer metastasis. The present disclosure also indicates that the concentration of succinate in the serum of lung cancer patients is higher than that of healthy subjects, which means that cancer cells release a large amount of succinate into the circulatory system during the development and progression of cancer. As shown in the present disclosure, serum succinate has high discriminative power and can be used as a new class of circulating metabolic carcinogen for predicting non-small cell lung cancer. In addition, in LLC tumor models, the increase in succinate concentration is accompanied by an increase in TAMs in subcutaneous tumors and more severe cancer metastasis, so the serum succinate may be an effective therapeutic biomarker for treating NSCLC. Therefore, the present disclosure also provides an anti-succinate antibody that can be used as a cancer therapeutic agent.
