Embodiment 1

[0028] 1. Preparation of Ethanol Extract (TPME):

[0029] Weigh 200g of yellow water branch coarse powder, add methanol 2000mL for the first time, soak for 1h, heat and reflux for extraction for 1h, add an equal volume of methanol for the second time, heat and reflux for extraction for 1h, combine the extracts, filter, take the filtrate, and recover the solvent under reduced pressure , obtained after freeze-drying.

[0030] 2. Establish CCl 4 Induced mouse model of liver fibrosis.

[0031] 2.1 Experimental animals and drugs

[0032] 2.1.1 Animals

[0033] Sixty male Kunming mice of SPF grade, weighing 18-22 g, were provided by the Experimental Animal Center of Guizhou Medical University. They were raised in a well-ventilated environment with a temperature of 18-25°C, a relative humidity of 40%-70%, and a 12-h light day and night cycle. License number SCXK (Qiang) 2018-0001. Relevant animal experiments were reviewed by the Experimental Animal Ethics Committee of Guizhou Medi...

Embodiment 2

[0042] Effect of TPME on liver index in mice with hepatic fibrosis

[0043] On the basis of Example 1, calculate the liver index, the formula is as follows:

[0044]

[0045] The results are shown in Table 1.

[0046] Table 1 Effect of TPME on liver index in mice with liver fibrosis

[0047]

[0048] Note: Compared with the blank group, * P** P# P## P<0.01

[0049] It can be seen from the experimental results that the liver index in the model group was significantly increased compared with the normal group, and the difference was statistically significant (P<0.01); the liver index in the colchicine group and each dose group of TPME were significantly decreased compared with the model group, and the difference was statistically significant (P<0.01). Statistically significant (P<0.01).

Embodiment 3

[0051] Effect of TPME on ALT and AST levels in serum of mice with hepatic fibrosis

[0052] On the basis of Example 1, the activities of ALT and AST in serum were determined by microplate method. The results are shown in Table 2.

Abstract

The invention belongs to the technical field of traditional Chinese medicine application, and particularly relates to application of a tiarella polyphylla alcohol extract in preparation of drugs for treating or protecting and regulating hepatic fibrosis diseases. The application comprises the following steps of: extracting tiarella polyphylla with methanol, heating and refluxing, adding methanol again for reflux extraction, combining extracting solutions, filtering, recovering a solvent under reduced pressure, and performing freeze drying to obtain the tiarella polyphylla alcohol extract. By establishing a mouse hepatic fibrosis model, the tiarella polyphylla alcohol extract is used for administration of high, medium and low doses of the mouse hepatic fibrosis model, and researches find that the tiarella polyphylla alcohol extract has a certain protective effect on CCl4-induced hepatic fibrosis mice, and the effect is related to inhibition of oxidative stress reaction, reduction of inflammatory factor level, regulation of TGF-beta 1/Smad signal pathway related protein expression, inhibition of HSC activation, reduction of alpha-SMA expression and reduction of collagen synthesis.

Description

technical field [0001] The invention belongs to the technical field of application of traditional Chinese medicines, and in particular relates to the application of ethanol extracts of Rhododendron japonicus in the preparation of medicines for treating, protecting and regulating liver fibrosis. Background technique [0002] Hepatic fibrosis (HF) refers to the pathophysiological process characterized by abnormal hyperplasia of connective tissue in the liver caused by chemical toxic injury, chronic hepatitis virus infection, autoimmune liver disease, alcoholism and other factors. The treatment of liver fibrosis by traditional Chinese medicine has the advantages of high curative effect, low toxicity and side effects, and low cost, and has gradually attracted people's attention. [0003] HF is a necessary pathological process for various chronic liver diseases to develop into liver cirrhosis, and its essence is the body's self-repair after liver tissue damage. The central link ...
