Embodiment

[0082] In addition to their well-characterized functions in tumor metastasis, the transmembrane glycoprotein podocalyxin-like protein 1 (PODXL, also known as podocalyxin-like protein 1, PCLP1, MEP21, Gp200 / GCTM-2, and thrombus adhesion protein) in human pluripotent stem cells (hPSCs) remains unclear. Here, we demonstrate that knockdown of podocalyxin-like protein 1 (PODXL) in undifferentiated human pluripotent stem cells (hPSCs) significantly inhibits the self-renewal capacity that currently prevents the expression of c-MYC and telomerase proteins. Notably, induction or reprogramming of induced pluripotent stem cells (iPSCs) as well as expanded pluripotent stem cells (EPSCs) was severely blocked upon knockdown of podocalyxin-like 1 (PODXL). Consistently, upregulation of podocalyxin-like protein 1 (PODXL) promoted the renewal of human pluripotent stem cells (hPSCs), enhanced the expression of c-MYC and telomerase, and promoted the activation of induced pluripotent stem cells (i...

Abstract

The present invention relates to a method for regulating potency of pluripotent stem cells (PSCs) by modulating expression of podocalyxin-like protein 1 (PODXL) and applications thereof.

Description

[0001] related application [0002] This application claims the benefit of U.S. Provisional Application No. 62 / 784,942, filed December 26, 2018, under 35 U.S.C § 119, the entire contents of which are incorporated herein by reference. technical field [0003] The present invention relates to a method and application for regulating the potential of pluripotent stem cells (pluripotent stem cells, PSCs) by regulating the expression of podocalyxin-like protein 1 (PODXL) and cholesterol. Background technique [0004] Human embryonic stem cells (hESCs) derived from the inner cell mass of early embryos have the ability to proliferate indefinitely and differentiate into endoderm, mesoderm, and ectoderm, and possibly into all cell types except the placenta (Thomson et al., 1998). Human embryonic stem cells (hESCs) behave like epithelial cells and are claimed to be primed (Brons et al., 2007; Kumari, 2016; Nichols and Smith, 2009; Tesar et al., 2007). Media switching can change primi...
