[0034] (1) Determination of cell transfection efficiency: In order to regulate the level of LINC01963 in lung cancer cells, short hairpin RNA (shLINC01963; 5'-CTCTCCCTACACCAGAATTTA-3') and LINC01963 overexpression plasmid were transfected into H1299 and A549 cells through Lipo 3000 middle. Empty vector was used as a negative control. After trypsinization of cells, cells were seeded at 2 x 105 cells / well in 24-well plates and grown to 70-90% confluency. Dilute Lipo 3000 (0.75 µL) and sample (1 µg) in Opti-MEM medium (25 µL x 2), then add 2 µL of P3000 reagent to the diluted sample. The sample-reagent mixture was then mixed with diluted Lipo 3000 (1:1) and incubated at room temperature for 10 minutes.
