Embodiment Construction

[0081] The present invention relates to the transgenic expression and purification of polypeptides from biological systems. More specifically, the invention relates to transgenic systems for expressing and purifying polypeptides, and methods of using these systems. The patents and publications cited herein reflect the prior knowledge of those skilled in the art and are hereby incorporated by reference in their entirety.

[0082] The present invention provides systems and methods for purifying target molecules present in biological systems. Systems and methods according to the invention utilize existing multivalent binding polypeptides, such as transgenically produced multivalent binding polypeptides, as affinity media to purify these target molecules. Preferably, the multivalent binding polypeptide binds both the target molecule and a matrix, such as a matrix capable of phase separation. These systems and methods provide many advantages over existing systems. The systems an...

Abstract

The present invention provides systems and methods for preparing and purifying target molecules present in biological systems. The systems and methods according to the present invention utilize a transgenically expressed multivalent binding polypeptide as an affinity medium for purifying the target molecule to purify the target molecule. The transgene multivalent binding polypeptide not only binds to the target molecule (for example, the bindable epitope of the target molecule), but also binds to the substrate.

Description

Background of the invention [0001] Preparation and purification of polypeptides for therapeutic and diagnostic purposes has become an important industry in recent years. Early attempts at recombinant polypeptide expression were performed in bacteria using plasmid expression vectors. For example, US Patent 4,816,397 discloses recombinant expression of immunoglobulins in bacteria. These methods are limited by the need to open or lyse the bacteria to obtain the recombinant polypeptide as part of a complex mixture containing bacterial proteins and other bacterial components. Yet another limitation is caused by the non-native oxidation state of recombinant polypeptides in bacteria, which leads to incorrect polypeptide folding. [0002] Some of these problems are alleviated by recombinant expression of polypeptides in mammalian cell culture fermentation systems. US Patent 5,639,640 discloses the recombinant expression of human follicle stimulating hormone (FSH) in mammalian cell ...
