Abstract

A Corynebacterium diphtheriae culture medium for the production of diphtheria toxin and methods for producing the toxin are provided. The medium is substantiallty free of animal-derived products and comprises water, a carbohydrate source, a nitrogen source and a number of free amino acids in an initial concentration wherein the initial concentration of each free amino acid is not limiting for the production of the toxin.

field of invention [0001] The invention relates to a bacterial growth medium for preparing diphtheria toxin and a preparation method of diphtheria toxin. Background of the invention [0002] Diphtheria is a life-threatening disease caused by infection with Corynebacterium diphtheriae, a Gram-positive, aerobic, rod-shaped bacterium. The disease is caused by a local attack of the nasopharyngeal tissue by a toxigenic strain of Corynebacterium diphtheriae. The organism grows within a rough fibrin membrane covering sore, hemorrhagic, and necrotic lesions, which may be located in the tonsil or nasopharyngeal region. During a typical epidemic in the past, the spread of the disease was through droplet transmission. Unless aggressively treated with antibiotics, a recovering diphtheria patient may harbor toxigenic bacteria in the larynx and nasopharynx for weeks or months.
