Abstract

The invention discloses a purifying method of rice bran protein through composite enzyme, which comprises the following steps: allocating 10%-20% slurry with degreased rice bran and water; adjusting pH value to 5.0-6.5 and temperature to 40-50 deg.c; building up phytase, hemicellulase, cellulase and aspergillus protein oryzae with rate at 15.64í† 15.06í†60.78í†8.52; adding 2-4% composite enzyme; stirring 1.5-2h; centrifuging 20 min; obtaining the supernatant as rice bran protein solution; condensing in the vacuum; freezing; drying; obtaining the product with protein receiving rate to 90.5-92.7%.

Example Embodiment

[0034] Example 1
[0035] 1. Weigh 100 g of defatted rice bran, add 900 mL of water, stir and disperse, prepare a slurry with a weight ratio of 10%, adjust the pH to 5.0-5.5 with HCl solution, and heat to 40±1°C.
[0036] 2. Add 0.31g of phytase, 0.3g of hemicellulase, 1.21g of cellulase, and 0.17g of aspergillus protease, mix well, enzymolyze in a 40±1℃ water bath, stir for 1.5 hours and adjust pH The value is maintained at 5.0 to 5.5.
[0037] 3. After 1.5 hours of enzymatic hydrolysis, terminate the reaction, centrifuge in a high-speed centrifuge for 20 minutes, centrifuge at 3000r/min, and collect the centrifuged liquid. The precipitate was first washed with 100 mL of water, and then with 200 mL of ethanol solution, the ethanol concentration was 80%, and then centrifuged, and the centrifugal liquid was combined.
[0038] 4. The centrifugal liquid was concentrated by evaporation under reduced pressure, and 12.2 g of rice bran protein was recovered by freeze drying. The degree of protein hydrolysis was 3.08%, and the extraction rate was 87.14%.

Example Embodiment

[0039] Example 2
[0040] 1. Weigh 150g of defatted rice bran, add 850mL of water, stir and disperse to prepare a slurry with a weight ratio of 15%, adjust the pH to 5.5-6.0 with HCl solution, and heat to 42±1°C.
[0041] 2. Add 0.47g of phytase, 0.45g of hemicellulase, 1.82g of cellulase, 0.26g of aspergillus protease, mix well, enzymolyze in a 42±1℃ water bath, stir for 1.5 hours, adjust pH The value is maintained at 5.5 to 6.0.
[0043] 4. The centrifugal liquid was concentrated by evaporation under reduced pressure and freeze-dried to recover 15.5 g of rice bran protein, with a protein hydrolysis degree of 3.76% and an extraction rate of 73.81%.

Example Embodiment

[0044] Example 3
[0045] 1. Weigh 200 g of defatted rice bran, add 800 mL of water, stir and disperse to prepare a slurry with a weight ratio of 20%, adjust the pH to 5.0-5.5 with HCl solution, and heat to 45±1°C.
[0046]2. Add 1.25g of phytase, 1.20g of hemicellulase, 4.86g of cellulase, and 0.68g of aspergillus protease, mix well, enzymolyze in a water bath at 45±1℃, stir for 2 hours, adjust pH The value is maintained at 5.0 to 5.5.
[0047] 3. After 2 hours of enzymolysis, terminate the reaction, centrifuge in a high-speed centrifuge for 20 minutes, centrifuge at 3000r/min, and collect the centrifuged liquid. The precipitate was first washed with 100 mL of water, and then with 200 mL of ethanol solution, the ethanol concentration was 80%, and then centrifuged, and the centrifugal liquid was combined.
[0048] 4. The centrifugal liquid was concentrated by evaporation under reduced pressure and freeze-dried to recover 23.78 g of rice bran protein, with a protein hydrolysis degree of 4.06% and an extraction rate of 84.93%.
