Embodiment Construction

[0049] The following examples will further illustrate the present invention in conjunction with the accompanying drawings. This embodiment is carried out on the premise of the technical solution of the present invention, and detailed implementation methods and processes are given, but the protection scope of the present invention is not limited to the following embodiments. For the experimental methods without specific conditions indicated in the following examples, the conventional conditions or the conditions suggested by the manufacturer are usually followed.

[0050] Step 1, the extraction of the total DNA of Streptomyces to be selected

[0051] Suspend 5 mg of mycelia with 500 μl of lysozyme solution and incubate at 37° C. until the cells become translucent. Add 50% volume of 2% SDS, mix and shake for about 1min until the viscosity of the solution drops significantly, then add 1 times the volume of neutral phenol / chloroform, mix and shake evenly, centrifuge at 12000rpm f...

Abstract

The invention relates to a new enramycin-producing strain and a high-yielding mutant strain thereof. In the invention, streptomyces obtained by separating marine environment is screened by using a primer designed by aiming at the conserved domains of lipopeptide antibiotics synthetic genes, and the new enramycin-producing strain of lipopeptide antibiotics is obtained through homology comparison. The invention also utilizes a ribosome engineering technology to breed mutant strains which has increased yield of enramycin. The invention provides a new strain source for fermenting and producing the enramycin, and the mutant strains can be used for industrial production.

Description

Technical field: [0001] The invention relates to a peptide antibiotic-producing bacterium in the technical field of biomedicine, in particular to a new enramycin-producing bacterium and a high-yield mutant strain thereof. Background technique: [0002] Enramycin, also known as enleamycin, enramycin, anlemycin, and duramycin, is a polypeptide antibiotic produced by the soil actinomycete Streptomyces fungicidious ATCC 21013. The drug was developed by Takeda Pharmaceutical Co., Ltd. in Japan in 1966, officially registered in Japan in 1974, and has since been registered and widely used in many countries. The technology was acquired by Schering-Plough Animal Health Products Co., Ltd. of the United States and used to produce feed premixes. [0003] Enramycin has strong antibacterial activity against Gram-positive bacteria such as Clostridium, Streptococcus, Staphylococcus, and Diplococcus pneumoniae, and has excellent growth-promoting and improving feed utilization. antibiotic g...
