[0107] Lyophilized formulation of humanized anti-IL-23p19 antibody.

[0108] Lyophilized formulations of humanized anti-IL-23p19 antibodies were prepared as follows. Table 3 provides the batch formula of the humanized anti-IL-23p19 antibody. The final concentration of humanized anti-IL-23p19 antibody was 25 mg / mL. This batch formulation can be used to prepare lyophilized 50 mg / vial units as described in Table 4 below. Uses polysorbate-80 from plant sources. Additional citric acid or sodium hydroxide can be added to adjust the pH to the desired value of about 4.8 (±0.2). Use pH 4.8 to reduce opalescence when antibodies are reconstituted from pH 5.5-5.6 citrate buffer. Use Sterile Water for Injection (WFI) to bring the ingredients to a final volume of 40 L. Of course, correspondingly smaller batches could be prepared by proportionally reducing the amounts listed in Table 3.

Background technique [0002] Interleukin-23 (IL-23) is a heterodimeric cytokine composed of 2 subunits (p19 unique to IL-23 and p40 shared with IL-12). The p19 subunit is structurally related to the p35 subunit of IL-6, granulocyte colony stimulating factor (G-CSF) and IL-12. IL-23 mediates signal transduction through binding to a heterodimeric receptor consisting of IL-23R and IL-12β1 shared with the IL-12 receptor. Numerous earlier studies demonstrated that the consequences of genetic defects in p40 (p40 knockout mice; p40KO mice) were more severe than those found in p35KO mice.
