Problems solved by technology

[0005] At present, domestic research on Bacillus subtilis mainly focuses on the screening of strains, isolation and identification, antibacterial effect, analysis of the chemical components of the fermentation liquid, and the production of general medium, but there are few reports on the fermentation and cultivation of Bacillus subtilis. There are fewer reports on the spore rate. The spore rate of the existing culture methods is generally about 80%, and its productivity is not ideal. People have been working hard to improve the spore rate.

Embodiment 1

[0020] A method for high-density cultivation of Bacillus subtilis with a high spore rate, which uses 5 L of Bacillus subtilis as a bacterial species, inoculates it into a fermentation medium for cultivation after activation, and adds a spore formation-promoting medium during the fermentation cultivation.

[0021] (1) Activation culture:

[0022] The medium used for activation is composed of the following raw materials: 200g of beef extract, 700g of peptone, 300g of glucose, 300g of sodium chloride, 750g of agar, and 47750g of distilled water.

[0023] The conditions of the activation culture are as follows: adjust the pH value to 7.0, the temperature is 36.5° C., and activate for 21 hours.

[0024] (2) Fermentation culture:

[0025] The activated Bacillus subtilis was inoculated into the fermentation medium for culture, and the sporulation-promoting medium was added at 9 hours after the fermentation culture started.

Abstract

The invention provides a method for the igh density culturing of Bacillus subtilis with a high spore formation rate. The method comprises the following steps: activating Bacillus subtilis as a strain, inoculating to a fermentation culturing medium, and culturing, wherein a spore generation culturing medium is added during the fermentation culturing. The Bacillus subtilis is an environmentally-friendly microbe which can be widely applied. The method provides necessary conditions for the improvement of the industrialized fermentation productivity of Bacillus subtilis, improvement of the effective rate of a product and prolongation of the retention period of the product by improving the formula of the culturing medium and the culturing conditions. The addition of the Bacillus spp production promoting medium substantially improves the spore formation rate of Bacillus subtilis to above 95%, and makes the spore formation rate about 15% higher than the spore formation rate of 80% in present common harvesting time, so the method substantially improves the productivity.

Description

technical field [0001] The invention relates to a method for cultivating Bacillus subtilis with high spore rate in high density. Background technique [0002] Bacillus subtilis (Bacillus subtilis) is a spore-forming G+ bacterium. The active substances such as subtilisin, polymyxin, nystatin, and gramicidin produced during the growth of the bacteria are effective against pathogenic bacteria or endogenous The opportunistic pathogenic bacteria of sexual infection have obvious inhibitory effect; quickly consume free oxygen in the environment, cause intestinal hypoxia, promote the growth of beneficial anaerobic bacteria, and produce organic acids such as lactic acid, reduce the pH value of the intestinal tract, and inhibit other Growth of pathogenic bacteria; stimulate the growth and development of animal immune organs, activate T and B lymphocytes, increase the level of immunoglobulin and antibodies, enhance cellular immunity and humoral immunity, and improve group immunity; can...
